Objective:Evaluate the analgesic effect, safety, and the role in the mechanism of analgesia of Radix Cynanchi Paniculati micron powder (MP),and Radix Stephaniae Tetrandrae MP to anus and enterology postoperative pain model.Methods:1.Use mouse-hot plate-model and mouse model of writhing comparing with blank group, aspirin group, tramadol hydrochloride control group, to observe somatic analgesic effect of the experimental drugs; 2.Establish a mouse model of postoperative pain of anus and enterology, taking Enzyme-linked immunosorbent assay (ELISA) to determin the content of prostaglandin E2 (PGE2) andβ-endorphin(β-EP) in mice serum, and to study the analgesic effect and and the analgesic mechanism of the experimental drugs.3. Observe drug toxicity and adverse reactions with acute skin and mucosa irritation experiment.Result:1、Both Radix Cynanchi Paniculati and Radix Stephaniae Tetrandrae MP in mouse-hot plate-model and mouse model of writhing have clear analgesic effect.2、Radix Cynanchi Paniculati MP is effective insufflation to the postoperative-wound surface analgesia,and its role considerably correspond to tramadol hydrochloride material external; Radix Stephaniae Tetrandrae MP also has analgesic effect, and the effect is lower.3、Radix Cynanchi Paniculati MP can improve (3-EP content and reduce PGE2 content in serum; and Radix Stephaniae Tetrandrae MP can only reduce PGE2 content.4、The present study find no mice in skin or mucosa stimulation from Radix Cynanchi Paniculati UP and Radix Stephaniae Tetrandrae UP.Conclusion:The original single analgesia herbs MP since 1949-Radix Cynanchi Paniculati and Radix Stephaniae Tetrandrae insufflation are safe and effective to the postoperative-wound surface analgesia, espesically Radix Cynanchi Paniculati MP.
