Silver Lai Soup For Dyspepsia Composite Influenza Virus Infection Mice Immune Mechanism Research

Background:Wind-warm lung-heat disease is caused by wind-heat pathogenic factor with the lung-heat clinical symptoms, such as fever, cough, chest pain,etc, which is an acute pulmonary inflammatory lesions. Traditionally, insufficient Zheng qi is considered as one of the vital internal factors for children recurrent respiratory tract infection (RRTI), but we found that Yangming internal heat also played an important role in the development process of RRTI,which might lead to a high incidence of RRTI. Traditional Chinese Medicine (TCM) will help to achieve a better outcome by clearing the lung and yangming heat based on the TCM treatment principle.Contents:Three parts were included in this study:theoretic elaboration, literature review and experimental study.1. Theoretic elaboration:In-depth explore the theory of "Yangming internal heat syndrome" and "Stagnation and heat syndrome in lung and Yangming" raised by my tutor Prof. Xiaohong Gu in terms of her long-term clinical practice experience and analyzing the relative ancient and modern literatures to interpret.2. Literature review:1) Retrospective summarize the implication,etiology,pathogenesis,and treatment of Wind-heat lung-heat disease in TCM knowledge.2) Retrospective summarize the relationship between children constitution and RRTI and the significance of conditioning constitution to prevent RRTI.3. Experimental study:Building the dyspepsia mice model on the basis of previous study, and then infected wi th influenza virus FM1nasally. Observing and recording the mice general state, volume diet and weight. And then examining lung tissue pathology image, lung index, thymus index, the level of sIgA in lung tissue homogenate and IgA.IgM,IgG in blood serum to get the impact of Yangming internal heat acting on virus infection mice on lung mucosa barrier, cytokines, humoral immune function, and evaluate the effect of TCM experience prescription "Yinlai Decoction" acting on the dyspepsia mice compound with influenza virus FM1on cytokines, humoral immunity and pulmonary mucosa barrier protection and try to identify the working mechanism.Results:1. Li terature study:Fewliterature has been publ ished about the development and innovation of the Wind-warm lung-heat disease research involved in internal cause,and treatment, etc.But more attention have been paid on the inner pathogenies and constitution of RRTI.2. Experimental study:1) Virus infected mice group(D2) and dyspepsia compositing virus infected mice group (M2) had more serious pulmonary inflammatory response and had higher lung index than the normal group (D1)(P<0.05),and the lung index in the dyspepsia compositing virus infected and treated by Yinlai decoction mice group(Y2) had been decreased significantly(P<0.05),thymus index had been increased significantly (P<0.05);the thymus index of dyspepsia mice group(Ml) had been decreased(P<0.05),and dyspepsia mice treated by Yinlai decoction group(Yl) had been increased significantly (P<0.05).2) SIgA in D2.M2.M1groups have been decreased significantly(P<0.05),SIgA in Y1and Y2groups have increased significantly (P<0.05). IgM in D2and Ml had been increased, in Y1had been decreased significantly (P<0.05);in M2had decreased significantly(P<0.05), in Y2had been increased significantly (P<0.05).Conclusions:1. In this study, Yangming internal heat is defined as the heat of gastric bowel state which is due to various reasons, encounter exogenous pathogenic factors or others, may lead to some symptoms,then show signs of disease called as yangming internal heat syndrome.If Yangming internal heat syndrome patient are invaded by exogenous pathogenic factors,then he may be diagnosed as "the stagnation heat in lung and Yangming","Yinlai decoction" may be an effective treatment.2. Yangming internal heat can influence the body’s immune function to make it easier to catch virus infection by reducing the level of SIgA,IgG,TNF-a and thymus index. Stagnation heat in lung and yangming can cause more serious lung damage in terms of the low level of SIgA, IL-10.TNF-a in lung tissue homogenate and IgG, IgM, IgA in blood serum.While "Yinlai decoction" may improve the body’s immune function by increasing the level of SIgA, TgM, and IL-10.
