1Purpose:The length of the survival of breast cancer patients often depends on the occurrence of liver and lung, and distant metastasis. This study, via the research on the highly metastatic breast cancer cells MDA-MB-435S in vivo, explores how stephania tetrandra and its active ingredient tetrandrine xenografts influence the transplanted tumors and pulmonary metastasis in nude mice, which lays the experimental foundation for clinical application.2Method:Established the animal models in nude mice of the breast cancer cells MDA-MB-435S transplanted tumor. Randomly divided24nude mice into model group, tetrandrine group, cyclophosphamide group and stephania tetrandra particles group (N=6) with drug intervention for28days.Observed and recorded the general condition of the mice and the volumes of the transplanted tumors during the administration. Killed the nude mice the next day after the end of drug treatment. Then weighed the transplanted tumors from each group and calculated the inhibitory rates of both tumors and pulmonary metastasis.Employed the immunohistochemical method to detect the changes of the vascular endothelial growth factor (VEGF) in tissue slice that induced angiogenic changes and the intratumoral microvessel density (MVD), to explore the mechanism of antimetastatic effect of tetrandrine on molecular level.3Result:According to the curve of tumor growth and the tumor inhibition rate, it can be seen that the growth of transplanted tumors from the model group is faster than other groups. It illustrates that the tetrandrine and stephania tetrandra particles are to some degree effective against the growth of estrogen receptor-negative MDA-MB-435S xenografts in nude mice but the difference was not statistically significant (P>0.05).Based on thecounting of pulmonary nodules and the calculation of inhibitory rates of pulmonary metastasis, both tetrandrine and stephania tetrandra particles are effective against the pulmonary metastasis of breast cancerand the difference is statistically significant (P <0.01). The inhibitory rates of pulmonary metastasis of tetrandrine and stephania tetrandra particles are respectively57.6%and42.4%. The measurement with immunohistochemical method reveals that both tetrandrine and stephania tetrandra particles can decrease the VEGF expression in xenograft tumor and lung tissue (P<0.01). And they can also effectively reduce the transplanted tumor MVD(P<0.05) and the difference is statistically significant.4Conclusion:The study, for the first time found that the tetrandrine and stephania tetrandra achieve the anti-angiogenesis by decreasing the transplanted tumor and pulmonary tissue expression of VEGF and reducing the intratumor microvessel density. To achieve its anti-angiogenic effect, provided the experimental basis for further exploration of the treatment of breast cancer recurrence and metastasis.
