Purpose:This study investigated “Yi Tang Kang†different course of STZ ratshemorheology in order to explore traditional Chinese medicine “Yi Tang Kangâ€on the mechanism of prevention and treatment of diabetic microangiopathy.Material and method:1. Weighing220-250g Wistar rats were randomlydivided into normal group,70, model200, model group rats by intraperitonealinjection of streptozotocin (STZ)55mg/kg, and the control group was injecteddose citratesodium buffer, measure blood glucose after72h to observe the ratsthe general situation and the type1diabetic rats into the mold rate.2. Modelgroup, the blood sugar were randomly divided into placebo group and Yi Tang Kanggroup, normal group was given saline4ml daily once gavage, the placebo groupwas given the4ml placebo once daily gavage group was given Yi Tang Kang groupYi TangKang4ml once daily by gavage. The experimental period was12months,10rats were sacrificed at3,6,9, and12months of the experiment blood rheologytesting.Results:1. Experimental results of STZ rat prepared model:The model group rats have different levels of reduced activity, increased dailywater intake, increased urine output, weight loss, diarrhea and othermanifestations, and there have been eight rats died, the mortality rate was4.0%.192surviving rats, the highest blood sugar level of33.8mmol/L, a minimumof6.9mmol/L,. There are23rats, fasting blood glucose levels less than16.7mmol/L, the modeling is not successful, be removed. The remaining171ratsfasting blood glucose levels are consistently greater than16.7mmol/L, forthe successful modeling.2. Hemorheology experimental results on the different course of STZ rats:2.1Baseline results: Yi Tang Kang group0months, the placebo group in0months, the normal group in0months, normal group in3months, the normal group in6months, the normal group in9months, the normal group in12months,hemorheological indicators are in line with positiveno significant differencebetween the state and the group.2.2Validity of results:The target of Whole blood viscosity (high shear), whole blood viscosity (lowshear), plasma viscosity, erythrocyte deformation index, hematocrit, fibrinogen,the two groups overall significant difference; erythrocyte aggregation index,the two groups as a wholeThere was no significant difference.Beginning of3month, two sets of whole blood viscosity (high shear) than beforetreatment began to have a significant difference in the placebo group in the6months after whole blood viscosity (high shear) was significantly higher thanthe benefits of sugar and Sport. Beginning of3month, two sets of whole bloodviscosity (low shear) than before treatment began to have a significantdifference between the placebo group three months after the whole blood viscosity(low shear) was significantly higher than the benefits of sugar and Sport group.Beginning of3month, two groups of plasma viscosity than before treatment, asignificant difference between the placebo group12months after plasmaviscosity was significantly higher than Yi Tang, Kang group. Beginning of3month,two groups of red blood cell deformability index than before treatment beganto have significant differences, six months after the placebo group, whole bloodviscosity (high shear) was significantly higher than the benefits of sugar andSport group. Beginning of3month, the two groups hematocrit than beforetreatment, a significant difference, three months after the placebo group,hematocrit was significantly higher than the benefits of sugar and Sportgroup.Since3months, two groups of fibrinogen than before treatment,significant differences, three months after the placebo group, fibrinogen wassignificantly higher than yi tang kang group. Conclusion:1. STZ rats hemorheology Yi Tang Kang on a different course had a better:.STZ rat whole blood viscosity (low cut), to improve the six month duration ofthe whole blood viscosity (high shear), hematocrit and fibrinogen generation;Yi Tang Kang can improve the course of three and12months duration ofplasmaviscosity and increase red blood cell deformability.2. Early Chinese medicine intervention can delay the rate of progress of diabeticmicrovascular disease, reflects characterized by the traditional Chinesemedicine "treating disease".3. Generalized estimating equations (GEE) can not only observe the effect ofintervention on the overall index change, but also from a specific point in timeto see the changes of each indicator. |
