| Purpose:This topic is based on theory of TCM acupuncture.By observingthe"Sancai thorn" and routine use of acupuncture and the treatmentsfor the analgesic effect of renal colic,to find the differences ofthe methods and the differences acupuncture analgesia renal coliceffects by animal experiments. We choose the pain threshold changingof the rats,the pathological changes of rats’kidneys and ureters,the serum calcium(Ca2+),urine calcium(Ca2+),renal tissues calcium(Ca2+)value, the changes of prostacyclin(PGI2) and thromboxane(TXA2) inrenal tissues, changes of Prostaglandin E2(PGE2) in spinal cord toobserve the object.Detect the differences of different acupuncturemethods on the analgesic effect of rats’renal colic,in order toevaluate the efficacy,and to establish the specificity of the bestanalgesic effect and the mechanism of acupuncture techniques.Throughthis experiment we could find a most comprehensive and most in-depth treatment of renal colic to reveal the role of acupuncture theoryand guidance clinical practice.Materials and methods:Divide the60male rats into five groups randomly according tothe weight:control group(group A),model group(group B),and useconventional acupuncture group(group C),"Sancai thorn" â… group(group D),"Sancai thorn"â…¡ group(group E).So there are12ratsin each group.In this study,the day before molding,test the painthreshold of rats,7days after model made, test the pain thresholdof rats again. Then (after the beginning of the official experiment8days)the treatment begins.During modeling process and thetreating,pay attention to spontaneous pain behavior ofrats,especially the changes in the situation.Selected point for group C is BL23(肾俞); group D is alsoBL23;group E is a-shi-point. From the day after the officialexperiment for8days, acupuncture at8am everyday.Group Celectroacupuncture once a day,regular pulse stimulation,15min for each time.Group D electroacupuncture BL23according to“god,human,landâ€three part respectively,5min for each part.GroupE is as same as Group D,espect selected the a-shi-point.Acupunctureonce a day for7days.After these7days test the pain threshold ofrats,the pathological changes of rats’kidneys and ureters, serumCa2+,urine Ca2+,,renal tissues Ca2+, PGI2and TXA2in renal tissues,PGE2in spinal cord.Result:1.The assaying result of the pain thresholdThe pain threshold before modeling: no differences betweendifferent groups(p>0.05);The changes of the pain threshold aftermodeling: Group Bã€Cã€Dã€E has a great diversity with A(p<0.01);Thereis no difference between Group Bã€Cã€Dã€E(pï¹¥0.05);The changes ofpain threshold fater treating:Group B and A have an obvious decrease,it has statistics significance(p﹤0.01),and Group Cã€Dã€E have nosignificant changes with A(pï¹¥0.05);Group C and B have greatdifferences(p﹤0.05),Dã€E each compares with Group B, the difference is obvious(p﹤0.01); Group D and E each compares with Group C,thedifference is obvious(p﹤0.05);Campared between D and E, thereisno statistics significance(pï¹¥0.05).2.The content and changes of secum Ca2+,urine Ca2+and Ca2+in thekidney tissue of each group of ratssecum (Ca2+): There is obvious difference between B and A(p﹤0.01). Group D and E each has differenc with C(p<0.05). Thereis no difference between D and E(p>0.05).PGE2in spinal cord: Group B and A has obviousdifference(p<0.01). Group C and A has difference(p<0.05). GroupD and E each has no difference with A(p>0.05). There isdifference between C and B(p<0.05). Group D and E each hasobvious difference with B(p<0.01). Group D and E each hasdifferenc with C(p<0.05). There is no difference between D andE(p>0.05).4.The pathological changes in the rats’ kidney and ureter ofdifferent groupsGroup A is normal rats, and there is no crystal and inflammatorycells in the ureter, the mesenchyme of kidney is uniform with noinflammatory cells invading, no congestion, no space expandation,and the inner wall of the ureter is smooth with no crystal and necrosistissue ecclasis. Group B has an obvious congestion in glomeruli and with obvious inflammatory cells invading, and there exists crystaldistribution in the ureter. The mesenchyme of kidney had obviousinflammatory cells invading and congestion with space expanding, andthe inner wall of the ureter is not smooth with much necrosis tissueecclasis. Group C has little congestion in glomeruli and littleinflammatory cells invading. The mesenchyme has little inflammatorycells invading and mild congestion but still with space expanding.Part of the innerwall of ureter in not smooth with little necrosistissue ecclasis. Group D has little inflammatory cells invading inglomeruli, and th congestion is obvious lightened. The kidneymesenchyme has an obvious easing of inflammatory cells invading andcongestion with no space expanding. The inner wall of ureter is smoothwith no necrosis tissue ecclasis. Generally, the glomeruli of GroupE have no inflammatory cells invading. The congestion is obviouslyeased. The kidney mesenchyme has an obvious easing of inflammatorycells and congestion with little space expanding. The inner wall ofureter is smoother with no necrosis tissue ecclasis. 5.The changes of evaluation on spontaneous pain action(According toAttal etc. Evaluation methods)After modeling, expect the blank group, each of the other grouphas spontaneous action after modeling with no difference onfrequency(p>0.05).After treatment,except the blank group(GroupA),the other four group of rats compared with each two of them onpain frequency. Group C,D,E each has an obvious difference with B.Group D and E each has difference with C(p<0.05). There is nodifference between D and E(p>0.05).Conclusion:1.Different acupuncture methods have effects on renal colic rats.2."Sancai thorn" method on renal colic of rat,analgesic effect isbetter than use conventional acupuncture method, be specific to theanalgesic effect.3. Mechanism of the analgesic effect "Sancai thornâ€depend onincreasing renal calcium oxalate stones in rats induced by renalcolic pain threshold,reducing inflammation and edema, PGI2and TXA2 synthesis in renal tissues,PGE2in spinal cord,Ca2+concentration inserum,and reducing Ca2+oncentration in urine,renal tissues Ca2+concentration,improving renal function.4. Acupuncture for renal colic of analgesic action has closerelationship with specific practices of acupuncture,but nearly norelationship with the selection on points. |
