Object:The aboveground resources of Rhubarb have a long history of using, rhubarb leaves and shoots are often eaten as food in civil. The development and utilization of one-second of biomass resource is of important significance in comprehensive utilization and environmental protection of Rhubarb. These studies on chemical constituents and extraction technology are helpful to look for alternative resources parts of underground of rhubarb.Method:Main biological resources of aboveground of rhubarb include stems and leaves(blade and Petiole), This article mainly does system research on the leaves of Rheum officinale Baill.The general idea is firstly study on the contents of anthraquinones in various parts of root and leaf for providing a basis for the study of aboveground parts. Secondly, a preliminary study on the chemical composition by system experiment, chemicals comparison of aboveground and underground parts are based on TLC and HPLC-DAD-ELSD, for trying to find generality and individuality of chemicals in differentparts, Then, we use column chromatography method to isolate some chemical components from it. At last, optimize the process of extacting effective constituents from leaves of Rheum officinale Baill by Plackett-Burman combined with CCD response surface methodology, and set up a method of multi-assay depending on one determination(QAMS) to detect the contents of the other anthraquinones and studya method for quality control of extracts from the aerial part of Rheum officinale(EFAPOR).Result:(1) We first adopt the study on different parts of Rheum officinale Baill The content of total anthraquinones and free anthraquinones inroot are higher than that in leaf, while the content of emodin、chrysophanol in leaf are far more higher than that in other components, these are0.05%and0.03%.(2)We first adopt the study on chemicals of Rheum offcinale Baill. Major chemicals differences in aboveground and underground parts of it are reflected in the ethyl acetate and n-butanol fraction we isolated8chemicals from aboveground part, Structure identification of5are Respectively emodin、chrysophanol、aloe-emodin、 physcione and β-sitosterol.The other3unique chemicals of leaves isolated from n-butanol fraction have not yet Identificated because of Lack ofexperimental conditions.(3) We first optimize the process of extacting effective constituents from leaves of Rheum officinale Baill by Plackett-Burman combinedwith CCD response surface methodology The optimization extractionof total anthraquinone is67.25%ethanol,90minutes for reflux per time,28.1fold of solvent and2times for extraction; The emodin is85%ethanol,90minutes for refluxper time,40fold of solvent and2times for extraction.(4) We first set up a method of multi-assay depending on one determination(Q AMS) to detect the contents of the other anthraquinones and study a method for quality control of extracts from the aerial part of Rheum officinale(EFAPOR). TLC and fingerprint can be usedto identify qualitativelythe chemical constituent of EFAPOR, Ultraviolet spectrophotometry can be used to control the quantity of the total anthraquinone, and the method of QAMS can be used to control the quantity of aloe-emodin、chrysophanol and physcioneConclusion:The aboveground part of Rhubarb can be used as alternative resources parts of underground of rhubarb. Studies on chemicalconstituents and extraction technology provide reference for development and utilizationare of aboveground parts. The method of QAMS can be used to control the quantity of aloe-emodin、 chrysophanol and physcione, the method is simple, accurate with a higher repeatability.
