Traditional Chinese medicine is our country medicine advantages and characteristics, it also important source of new drug effective development. However, because of the complexity of the traditional Chinese medicine and clinical medicine flexibility so that the active ingredients of the new drug development constraints become screening process of bottleneck. Therefore, Chinese medicine complex system of drug in active ingredient efficient method to set up and application of screening will be in the medical field research focus of the eternal. In this thesis, we choose the cough medicine Semen Brassicae, which used widely in traditional Chinese medicine clinical,to conduct a systematic study of high-throughput screening its active ingredients. Exploring the methodology of the active ingredients of traditional Chinese medicine screening with the receptor chromatography. Designing to provide ideas for the receptor chromatography to screening of the active ingredients in traditional Chinese medicine. This paper is divided into four chapters, the author’s main contributions are as follows:1. Established high performance liquid chromatography method to detect sinapine of Semen Brassicae; Using orthogonal design method and response surface methodology to study the sinapine in the best extraction process. The results showed that the peak area and the concentration of sinapine gave a linear relationship during the range of0.0155~0.2945mg/mL,precision determination of the relative standard deviation is0.42%, reproducible determination of the relative standard deviation is0.61%, the average recovery is95.5%. It demonstrated that this method can be used to determining sinapine in Semen Brassicae.The best extraction method of sinapine in Semen Brassicae is hot reflux method, the best process is40times the amount65%ethanol60℃hot reflux extraction5h. It can be applied to use β2-adrenergic receptor chromatography to screening the active ingredients of Semen Brassicae and provide stable and reliable sample preparation and quality control methods. 2. The bioactive compounds of Semen Brassicae were screened by β2-adrenoceptor biochromatography,use the competitive dispalacement analysis method to study the interaction between receptor and the active ingredients.The result showed that sinapine is the active ingredient in Semen Brassicae which had the specific interaction with β2-AR, The association constant of sinapine with β2-AR in the biochromatographic column was1.36×105M-1, and the binding sites were1.27×10-6M. It proved that the receptor chromatography can be used for rapid screening of specific active ingredients in traditional Chinese medicine and its receptor interaction mechanism.
