Study On Preparation And Preliminary Pharmacodynamics Of Extract Of Modified Pipaqingfei Capsule

Ache is a chronic inflammatory dermatosis happened.in the sebaceous follicles. Its incidence is high,if not being treated promptly,it will take serious physical and psychological consequences to patients.Because the pathogenesis of ance is complex, western medicine has its limitation,and also generally follows many adverse effects such as irritation to the skin toxicity of genital system or drug resistance.It is a long history for traditional Chinese medicine to treat acne,curing it according to different disease type such as Feijingfengre Zheng,Piweishire Zheng and so on.Pipaqingfei decoction has been used to treat acne vulgaris.But up to now,the easily oral preparation of Chinese crude drug is still blank in the market.On the basis of the classic prescription Pipaqingfei decoction,the research group has prepared the modified Pipaqingfei capsule to treat acne vulgaris,adding many kinds of Chinese crude drug according to modern clinical experience.The research on the preparation technology and rudiment pharmacodynamics of the drugs was carried out in this thesis.The main contents and conclusions are as follow:1.According to the related literature,the classic prescription’s function and property of every medical material,extraction and purification technologies were designed.Using total flavones yield and total triterpenes yield as technology indexes,the single factor and RSM experiments were carried out for the extraction and purification of the group of Folium Eriobotryae,Cortex Mori,Radix Glycyrrhizae, Fructus Crataegi and Radix Codonopsis.The optimal technology is Chinese crude drugs weighed according to the proportion of the prescription are extracted three times with 16.5 times of 66%alcohol,2.0 hours each time.The pH of extract is adjusted to 4.0.The acidifying extract is diluted with 1.5 times water of pH 4,then centrifuged with 5000 rpm.The supematant is acidified to pH 1.0,then centrifuged with 5000 rpm.Both of the centrifuged preciptitate is washed colourless with water and put into the vacuum drying oven to get intermediate-Ⅰ.The yield of total flavones is 6.4 mg/g,the yield of total triterpenic acid is 32.1 mg/g,and the content of extractum is 64.9%.Using total alkaloids yield as technology index,the single factor and RSM experiments were carried out for the extraction of the group of Rhizoma Coptidis and Cortex Phellodendri Amurensis.The optimal technology is Chinese crude drugs weighed according to the proportion of the prescription are extracted 4 times with 18.125 times of water,1 hours each time.The pH of extract is adjusted to 12.The alkalizing extract is extractd with 1 times of acetic ether for 6 times.Both of the centrifuged preciptitate is put into the vacuum drying oven to get intermediate-Ⅱ.The yield of total alkaloid is 43.5 mg/g,and the content of extractum is 66.1%.2.Rudiment pharmacodynamics of modified Pipaqingfei was carried out in vitro. The drug was discussed by the assay of the tyrosinase inhibition,and was further investigated by anti-dopachrome autoxidation activity,free-hydroxy radical activity, anti-DOPA autoxidation activity,anti-ultraviolet catalysis activity,and promoting degradation of melanin.The cell culture discovers that the drug can inhibit the proliferation of B16 melanocyte.All these results demonstrate that the drug might have multi-target of inhibiting the synthesis of melanin and improve pigmentation effectively.3.The animal experiment indicates that the modified Pipaqingfei has the effect of improving the rabbit-ears hair follicles funnel keratosis level,promoting the generation of new hair follicles,reducing androgen levels,improving blood circulation especially for local microcirculation,treating inflammation,repairing skin damage,reducing blood lipid and lowering cholesterol.4.By testing,it’s reasonable to add Fructus Crataegi to Pipaqinfei.To fit clinical requirement,modified Pipaqinfei can reduce blood lipid and lower cholesterol.
