Objective:Differential proteomics were used to detect different proteins in borderlineovarian serous cystadenoma(IN)and cystadenoma(BE) in this study. In order to findspecific biomarkers, we combined bioinformatics method with comparative analysis,Screening out the differences between the two proteins. This experiment screened36different proteins. Afterfurther experimental analysis and clinical verification, it willbecome a tumor markers of borderline ovarian serous cystadenoma, and haveimportant significances in pathogenesis, diagnosis, treatment and Prognostication forovariantumors.Methods:Two mixed samples were extracted from IN and BE tissue samples in this study.Two independent experiments were done using two dimensional liquidchromatography MS/MS spectrometry analysis, Differential proteins were obtainedby comprehensively and comparatively analyzing the results of twoexperiments,ultimately. According to the determination principles, Distinguishing thedifferential proteins between IN and BE.Results:1.By Using two dimensional liquid chromatography MS/MS spectrometryanalysis technology,585proteins were detected from borderline ovarian serouscystadenoma group,and381proteins were identified in ovarian serouscystadenoma.Finally,36differential proteins were identified from the two proteinsaccording to principles. There are31proteins low-expressed and5proteinsover-expressed in ovarian serous adenocarcinoma.2.The diffential proteins mainly including the enzymes involved in the materialenergy metabolic,extracellular matrix proteins, cytoskeletal proteins,the chaperone protein, tumor nucleic acid metabolism related proteins,and proteins involved in cellcycle, apoptosis, transcription and translation, DNA repair genes, tumorimmunity-related factors,and other proteins involved in ovarian cancer occurrence,development, and metastasis-associated protein.Conclusion1.The method is so efficient,sensitive and repeatability. Diffential proteinspectrum of borderline ovarian serous cystadenoma was preliminarily built by usingtwo dimensional liquid chromatography MS/MS spectrometry analysis technique todetect tissue samples and screen proteins from IN and BE tissues.2.In this study a total of36differential proteins were identified from IN andBE.Some proteins’abnormal expression in IN and BE has never been reported,andmany of these proteins will be beneficial in the differential diagnosis in IN and BE.3. Further retrieving the differential proteins found that the nzymes involved inthe material energy metabolic,extracellular matrix proteins, cytoskeletal proteins andproteins involved in cell cycle, apoptosis, transcription and translation play moreimportant roles in carcinogenesis、development and metastasis of borderline ovarianserous cystadenoma.4.Some proteins associated with tumor abnormal material and energymetabolism may be helpful in enriching the metabonomics of ovarian tumors.5. The experimental results will be compared with the other five groups, andfinally the sifted proteins may help to perfect the existing ovarian cancer proteomedatabase. Ultimately we will identify valuable tumor markers for early diagnosis ofovarian cancer.
