The lack of specific clinical symptoms and signs of early stage ovarian cancer and effective early diagnostic techniques,70% of patients when clinical treatment late. Looking for a way to early diagnosis of ovarian cancer with high sensitivity and high specificity is an urgent problem which need to be solved as quickly as possible.With the completion of the human genome project, bio-science technology stepped into the post-genome era. Proteomics as a new area of research in post-genome era is used to research large quantities of clinical specimens, such as serum, organizations and do a lot efficient proteome analysis, which makes it possible to seek for specific and sensitive biomarkers in early clinical diagnosis.Proteomics is different from the traditional disciplines of protein, and its research was conducted in the whole organism or its cells on the protein level, which reveal the basic law of the overall activities and clarify the activities of life from the perspective of an organism or a cell protein. Proteomics is the study of all proteins within cells, the composition and dynamics of an emerging discipline. The rapid development of its cancer research and finding new tumor markers provides a new way of thinking and technology platform.Objective:In this study, application of proteomics to detect different proteins in ovarian serous adenocarcinoma and ovarian serous cystadenoma, further integration method of comparative analysis of biological information, from which the difference between selected proteins in order to find specific biomarkers. This study is only one part of proteomics experiments, and will compared with the other five groups and finally identify some ovarian cancer-specific tumor markers for the early diagnosisMethods:In this study, ovarian serous adenocarcinoma and ovarian serous cystadenoma tissue samples obtained by two mixed samples, using two-dimensional liquid chromatography-mass spectrometry analysis conducted two experiments, a comprehensive two experimental results, ultimately, ovary serous adenocarcinoma, ovarian serous cystadenoma tissue differences in the respective proteins, and then according to principle of differential proteins to identify ovarian serous carcinoma and ovarian serous cystadenoma.Results:1. By Using two-dimensional liquid chromatography-mass spectrometry analysis technology, a total of 1085 proteins was identified from ovarian serous adenocarcinoma group, and a total of 381 proteins was identified from ovarian serous cystadenoma group According to principle of different proteins, a total of 128 differential proteins were identified from ovarian serous adenocarcinoma and ovarian serous cystadenoma. there are 116 proteins over-expressed in ovarian serous adenocarcinoma, and 12 proteins low-expressed in ovarian serous adenocarcinoma. The following proteins are low-expressed:Lumican, collagen a-3 (â…¥) 2,3,4 precursors, collagen XIV-A1, desmin, histone H1 1-D chain, POTEE, decorin, fibromodulin, PRELP, Osteoglycin.2. the diffential proteins mainly include the extracellular matrix, cytoskeleton components, tumor immunity-related proteins, and tumor metabolism related proteins, molecular chaperones, and other proteins involved in ovarian cancer occurrence, development, invasion and metastasis-related.Conclusion:1. In this study, we identified many differential proteins from ovarian serous cystadenocarcinoma and ovarian serous cystadenoma, many of which are reported to be abnormal expressed in ovarian serous adenocarcinoma for the first time. These proteins will be beneficial in the differential diagnosis of ovarian serous cystadenocarcinoma and ovarian serous cystadenoma. The experimental results will be compared with the other five groups, and finally some valuable tumor markers will be identified, which is useful for the early diagnosis of ovarian cancer2. the proteins which are related to the abnormal material and energy metabolism can be used for the modeling used for diagnosis of ovarian cancer.
