| Objective:To investigate the effects of the WenYang ZhenShuai(WYZS) granule on the rabbit model of chronic heart failure induced by Adriamycin and its impact on ERK5expression.Methods:60rabbits were randomly selected15for normal group, the remaining45were given Adriamycin by intravenous injection for4weeks. After4weeks, all rabbits were did echocardiography.13rabbits were selected for4weeks model group randomly, and we removed their hearts to detection and quantitative T-ERK5, p-ERK5and quantitative of ERK5mRNA expression. Then the30rabbits which were given Adriamycin were assigned randomly into5groups by the heart function: Chronic heart failure (CHF) model group, positive control group, WYZS granule low-dose group,WYZS granule middle-dose group,WYZS granule high-dose group. In addition to the normal group injected with saline, the other5groups were injected with Adriamycin to the8th week. On the5th week, the normal group and CHF model group were given saline, all of the WYZS granule groups were given WYZS granule infusion, the positive control group were given enalaprilat for4weeks. After experiment, all rabbits were did echocardiography and be removed their hearts to detection and quantitative T-ERK5, p-ERK5and quantitative of ERK5mRNA expression.Results:1. Compared with normal group, the cardiac function in4weeks model group and CHF model group decreased significantly (P<0.01), the cardiac function of positive control group and WYZS granule group were improved, compared with CHF model group, the difference was significant (P<0.05). Compared with positive control group and WYZS granule group, the cardiac function of WYZS granule middle-dose and high-dose group was better(P<0.05). The differences between positive control group and WYZS granule low-dose group, WYZS granule middle-dose and high-dose group were not significant (P>0.05).2. The differences of T-ERK5among all groups were not significant (P>0.05). Compared with normal group, p-ERK5of the other group were decreased (P<0.05). But positive control group and WYZS granule groups’p-ERK5were increased compared with CHF model group(P<0.05). The differences between positive control group and WYZS granule high-dose group, WYZS granule low-dose and middle-dose group were not significant (P>0.05). The differences of ERK5mRNA among all groups were not significant (P>0.05). Conclusion:WYZS granule can improve heart function on rabbit model of chronic heart failure induced by Adriamycin. And middle-dose and high-dose of WYZS granule were better than WYZS granule low-dose and enalaprilat. The effects of WYZS granule may realized by means of increasing p-ERK5to regulate ERK5signal transduction pathway. But the specific mechanism needs further study. |
