| Pleurotus cornucopiae, delicious, nutrient-rich, is our daily consumption favored large edible fugus which has been promotion cultivation feed in many places of our cou ntry. It contains protein, amino acid, fat, minerals, vitamins,iron,calcium and other n utritional ingredients.Recent studies suggest that edible fugus can inhibit tumor growth, fall blood sugar, have antioxidation, antivirus and regulating the immune functions. It sh ows great application prospect in medicine and functional food fields.In this dissertati on, research of purification, physicochemical properties and otective effects of it on acut e hepatic injury in mice.1Isolation and purification of polysaccharide of pleurotus cornucopiae, and appraisal of their physicochemical properties. A crude polysaccharide(PCNP I) from pleurotus cornucopiae was obtained by boiling water extraction, ethanol precipitation. The refined polysaccharides(PCNP II) were obtained after removal of protein by Sevga method and drying by drier, followed by consecutive purification on DEAE--Cellulose52column and Sepharose4B column,the PCNP Ⅱ was purified to a homogeneous polysaccharide(PCNPⅢ).The purity of PCNPⅢ powder was more than94.37%.Taking vitamin C as contrast, studies the antioxidant of PCNP Ⅱ and PCNPⅢ.It found that when The purity of PCNPⅢ was more than90%,indicating that it has a effective scav enging of free hydroxyl radical(·OH),and its scavenging rate close to90%of Vc. But the scavenging effect of O2-is65%of Vc.2Taking refined polysaccharides(PCNP Ⅱ) as the test material and establish acute alcoholic hepatic injury model to investigated the protective effect of PCNP II in mice(ACL). The mice were administered by gavage for30days prior to the administrati on with dose of50%alcohol (12mL/kg.BW) except blank control group.12hours after the last alcohol treament, all mice were dislocation death to assay activities of antioxidant capabilities in blood and liver. Results showed that ACL-PCNP Ⅱ-M group serum activites of AST, ALT, TG were obviously improved; compared with the model group, the contents of GSH and activities of SOD,CAT, in the PCNP Ⅱ groups liver we re increased, the contents of MDA in the liver were decreased, obviously in the ACL-P CNP Ⅱ-H treatment groups(P<0.01).3The further determine of the the protective effect of PCNP Ⅱ in mice is to establish acute CCl4hepatic injury model. The mice were administered by gavage for30days prior to the administration with dose of CCl4(80mg/kg.BW) except blank control group.At24h postadministration, all mice were sacrificed to assay activities of antioxida nt capabilities in blood and liver. Results showed that serum activities of AST, ALT, ALP were decreased to the CCl4model group lever after PCNP Ⅱ administration,but the mice were still liver injury,also the contents of GSH and activities of SOD and CAT increased in the liver,but approximate BIFENDATE group,decrease of MDA.4Effects of PCNPⅡon microstructure and ultrastructure of injured liver of mice by alcohol or CCl4Pathological microscopic structure shows that a large number of short-term drinking can cause hepatocyte swelling, shrinkage and deformation,and the CCl4model group was serious,whereas the PCNP Ⅱ treated groups relieved the liver cell damage. The further observation of ultrastructure supported the conclusion of microstructure. |
