ObjectiveExplore the influence of nerve growth factor gene transfection ofperiodontal ligament cell osteogenic, evaluation of the possibility of seed cellsfor periodontal tissue regeneration and the role of promote periodontal tissueregeneration.MethodsNGF gene was transfected into human periodontal ligament cells throughthe pcDNA3.1-NGF by using the gene transfection techniques. Identify theexpression of transfected cells by using the SP method ofimmunohistochemistry and western blot and observe the impact of osteogenic. The experimental specimens are the orthodontic removal of the maxillarypremolar specimens from Liaoning Medical University Second AffiliatedHospital, cryopreservation at-70°C. Use SPSS13.0software package forstatistical analysis.Result1. Immunohistochemistry confirmed that the NGF protein was expressedin periodontal ligament cells after the NGF transfection.2. Immunohistochemical staining showed positive reaction to anti-Bosiantibody, anti-keratin antibodies were negative, to prove that cells derived frommesoderm.3. the contents of OC in PDLC after the transfection of NGF group is morethan the untransfected NGF group(P<0.01).4. Western blot showed that transfected with the NGF group wassignificantly increased, and the exogenous DNA does not affect the cells themselves NGF synthesis.ConclusionPeriodontal ligament fibroblasts cells transfected with NGF gene cansignificantly enhance the cells’ ability of osteoblasts. In this study we transfectthe NGF gene into vitro PDLC, analysis of cell osteogenic, made it possible forperiodontal ligament cells as seed cells of the periodontal tissueregeneration.Lay the foundation for further study of the combination of cellbiology and tissue engineering for the repair of periodontal tissue defect. |
