There has about120species of the Annona genus in family Annonaceae, mainly distributing in tropical America, a few in tropical Africa. The6kinds of Annona existing in our country are introduced from tropical America. This genus contains polyketides, terpenes, cyclic peptides, alkaloids and other ingredients, which has strong biological activity in antitumor, antimalarial, antibacterial, immune suppression, poor feeding, insecticidal and so on.Annona densicoma Mart, is located in the Amazon rainforest of South America, which the natives called graviola-brava, often used as a folk medicine. Eight polyketides ingredients have been isolated from Annona densicoma Mart.; the alcohol extract of bark and Annonaceous acetogenins have been proved strong inhibitory activity in vitro tests against P388, A549, HT29and other cell tumor lines.In order to further development and utilization of plant resources of Annona densicoma Mart., this subject studied the chemical constituents of Annona densicoma Mart, stems collected from Peru, by using normal phase, reverse phase silica gel column chromatography, MCI Resin column chromatography, Sephadex LH-20column chromatography, HPLC and other Modern isolation technique, I made a aystematic study on Anti-tumor activity, that was found in dichlormethane extract and separated and obtained18compounds, two of which are flavonoid:quercetin (AD-12), and eriodictyol (AD-13), three are organic acid:protocatechuic acid (AD-11), syringic acid(AD-10) and Stearic acid (AD-2), one is amide alkaloids: N-p-hydroxy-cis-coumaroyltyramine(AD-14), two are polyketides:annonacin(AD-4), annonacin-10-one (AD-5) and β-sitosterol (AD-3) and daucosterol (AD-1). Besides, it is the first time that protocatechuic acid was separated from Annona, compound AD-1,AD-2,AD-3,AD-10,AD-12,AD-13and AD-14was separated from Annona densicoma. Mart.The rest of compound is under structural dentification, and can be inferred to be polyketides from the1H-NMR and13C-NMR data.Besides the study above, we also pilot study on vitro cytotoxic activity screening of extractive. It is found that dichlormethane extract F01of Annona densicoma Mart, crude extraction has a strong inhibition activity in A549, Belã€A875, the IC50of which is33.839μg/mL,30.542μg/mL,59.455μg/mL. |
