Annona glabra Linn, Which is also called Annona glabra, is one plant of Family Annonaceae and Annona Genus. This plant is native to tropical America, and has already been introduced in provinces of Guangdong, Guangxi, Yunnan, Zhejiang, Hainan, Taiwan, and Hong Kong in China. It has been used as an insecticide and insect repellent in traditional Chinese medicine for a long time. The first study of Annona glabra Linn. was dated from the late1980s, which had isolated three adjacent-bis-THF AAGs from it:squamocin, asimicin and desacetyluvaricin. And others separated some compounds from the parts of bark, seed, fruit, such as terpenoids, cyclic peptide, alkaloids, which have different kinds of biological activities. Especially, the antitumor activity of the lannonaceous acetogenins (AAGs) is remarkable to human society.This article focuses on the seeds of Annona glabra Linn. which were farmed from Guangdong province of China, uses the cell culture technique in vitro to screen the parts of antitumor activity in the seeds crude extract of Annona glabra Linn, based on the existed research foundation. The SRB method was used to initial screen the pharmacological activity of seeds crude extract worked on the cytotoxin F06and F07, calculating the IC50value of inhibiting tumour cell growth, and ensuring the sensitive areas. The result showed that the cytotoxin activeness of F06cells was superior to F07part, the IC50values worked on Bel-7402(human hepatoma cell line), HCT-8(human colorectal carcinoma cell line) HEPG-2(human liver cell line) were53.63μg/ml,82.73μg/ml, and89.47μg/ml, respectively.The sensitive part which was screened by cell culture was F06.Thirteen compounds were isolated by the means of the routine, medium voltage silica gel column, Sephadex LH-20column chromatography, HPLC preparation and many kinds of modem separation technologies, and seven compouds structure were identified through the classic chemical methods and various advanced spectrum learning techniques (including1H-NMR,13C-NMR, HMBC, HMQC, EI-MS, etc). Six componds among which were lactone compounds:Desacetyluvaricin (AG1),4-deoxyasimicin (AG2), Asimicin (AG3), Bullatacin (AG4), Motrilin (AG5),4-desoxycherimolin-2(AG8), and sucrose (AG9). AG9was isolated from this plant for the first time, and the rest of the compounds need further identification. By analyzing the data of1H-NMR and13C-NMR, there were five lactone compounds, one alkaloids compound.
