Objective:This project is intended to establish an animal model which with allergic rhinitis Syndrome on spleen deficiency rats, Explore the effect of the Wulong granule to spleen deficiency in rats with allergic rhinitis local and systemic, providing the experimental basis for the clinical application of Chinese medicine treatment of allergic rhinitis.Method:Making Spleen animal models, according to Qiu Baoshan reported rhubarb bitter cold diarrhea dismount to observe rhubarb on body temperature and weight of the rat. Copy the animal model of allergic rhinitis intraperitoneal injection of ovalbumin sensitized. Exciting the nose locally at the same time giving Wulong granule in the high, medium and low-dose gavage treatment through intragastric infusion, and establishing control group (normal feeding gavage), model group (daily with the drug group tolerance of normal saline), the comparison group (loratadine orally). To observe the the Wulong granule on the Spleen allergic rhinitis rat body temperature, weight, nasal symptoms, nasal secretion, using the standard swatches measuring nasal septum mucosa pH values, after continuous medication for7days. ELISA was used for the detection of IL-5in rats. Light microscopy combined with graphic image analysis system to observe the inflammatory cell infiltration of the nasal septum mucosa, eosinophil count, mastcell count and degranulation of the situation. Results:Weight of spleen deficiency with allergic rhinitis rats is slow growth, hypothermia, poor general condition; pH values of nasal secretions is lower, local environment is acidic; the IL-5levels is higher; the nasal septum mucosa is more fragile, inflammatory cell infiltration is more severe, eosinophils, mast cells are increased. The above-mentioned pathological conditions, high doses Wulong granule to improve the role of high dose significantly. Conclusions:The Wulong granule can reduce the infiltration of inflammatory cells, by regulating the expression of cytokines IL-5. To play a therapeutic role of rats with experimental AR. |
