Objective: To observe the effects of20(s)-protopanaxadiol (PPD) onproliferation and apoptosis of Siha cells in vitro and clarify the mechanism of thoseeffects.Method Siha cells cultivated in vitro were treated with alcohol and differentconcentration of PPD respectively. Phase contrast microscope was used to observeCell morphology of treated Siha cells. MTT was used to detect the effect of PPD onSiha cells proliferation, AnnexinV-EGFP/PI double staining and TUNEL stainingwas used to detect DNA damage of treated cells, flow cytometry was used to detecteffect of PPD on cell cycle of Siha cells. The transcription and protein expression ofp53, p21, cyclinE, Bax, caspase-9and caspase-3were analyaed by RT-PCR, Westernblot and immunocytochemical staining.Result Compared with negative control group, after treated with differentconcentration of PPD,1. Changes of cell morphology of treated Siha cells wereobserved;2. MTT showed PPD could inhibit proliferation of Siha cells;3.AnnexinV-FITC/PI and TUNEL staining results showed PPD would induceapoptosis of Siha cells;4. flow cytometry results showed PPD would induce Sihacells G1/S cycle arrest;5. the transcription and protein expression of Bax、Caspaes9、Caspaes3、p53and p21were up-regulated, while the transcription andprotein expression of cyclinE was down-regulated.Conclusion20(s)-protopanaxdiol (PPD) may suppress the proliferation ofSiha cells, and induce the G1/S cycle arrest. The mechanism may be related toup-regulation of P53and P21, and down-regultion of cyclin E. PPD may induce apoptosis of Siha cells, the mechanism may be related to activition of Caspaes3signaling pathway.
