| Objective:Establishing guinea pigs anaphylactic shock model.and making specimens of blood serum. Elisa method to detect the expression of guinea pig serum IgE and IL-10,Trying to provide objective diadynamic criteria to diagnose anaphylactic shock for forensic. Methods:25healthy adult guinea pigs of either sex, weighing about200g, classified into the experimental group of15and the control group fo10with no purpose. The experimental group were immunized intracutaneously with human mixed serum and then induced by endocardiac injection after3weeks. The control group injected with saline and Put to death by roken cervical spinal cord. Clinical manifestations observed after the re-injection of the guinea pig. Perform autopsies on two groups of guinea pigs. Blood was drawn from heart and the serum was purified. Larynx, trachea, lungs, and stomach of guinea pigs were taken out, then fixed in4%polyoxymethylene and embedded in paraffin. Elisa method to detect the expression of guinea pig serum IgE and IL-10, SPSS18.0software to analyze experimental data. Results:(1) Experimental group, all the typical clinical manifestations of anaphylactic shock in again after the injection within1min and all death within2-30minutes; Control group did not change.(2) The guinea pigs larynx and trachea were of edema, pulmonary, alveolus were of expansion, and alveolar wall becamethin with HE staining in anaphylactic shock; Degranulation of mast cells in lung tissue was observed.(3) The IgE levels of serum in experimental group is higher than that in control group, P<0.05;(4) The IL-10levels of serum in experimental group is higher than that in control group, P<0.05. Conclusion:C1) Successfully established a guinea pig in anaphylactic shock animal models.(2) Anaphylactic shock of guinea pig serum IgE and IL-10levels were significantly increased.(3) ELISA measurement of serum IgE and IL-10can assist in diagnosis of anaphylactic shock. |
