Ampelopsis mega lophylly Diels et.Gilg, a medicinal plant of Ampelopsis, is a herb commonly used in the west area of Hubei province, which is neutral in nature and has bitter and sour tasting. The preliminary research of this subject contains three parts. First, studies on chemical constituents, ten compounds were isolated and identified from this plant. Seconds, studies on quality standard, the Chromatographic Fingerprints of the effective fraction obtained from ten batches of this medicinal materials, which were collected from different places, were established. Finally, pharmacological study shows that Ampelopsis megalophylly have antihypertensive effect and anti-HBV activity in vitro.Additionally, we established high performance capillary electrophoresis fingerprint of the extract, and researched the effects of this extract and the ampelopsin on lowering blood pressure to preliminarily discuss its mechanism. The content of total flavonoids was determined with ultraviolet spectrophotometery(US) and ampelopsis with high performance liquid chromatography (HPLC) respectively. The precise method to identify ampelopsis and myricetin with TLC was established. Limit test mainly consists of Arsenic salt, Heavy metals, Pesticide residues, loss on drying etc so as to conduct a overall effective control of the inner quality of the total flavonoids.Spontaneously Hypertensive Rats (SHR) were used as pharmacological model to observe the influence of the different content of the total flavonoids and the ampelopsis on the blood pressure, Angiotensin â…¡ (Ang â…¡) as well as Endothelin (ET) of SHR, and also to investigate the mechanisms of lowering blood pressure. The above results show that the total flavonoids and the ampelopsis have significant effect on lowering blood pressure of SHR at different degree, and yet, did not significantly reduce the plasma Ang â…¡ and ET content in the plasma. So mechanism of action remains to be further researches.
