Objective:To explore the influence of alendronate on fracture healing of rat’s osteoporosis fracture.Methods:25adult female SD rats were randomly divided into two groups:20for ovariectomized group,5for blank control group.6weeks later from ovariectomy in the ovariectomized group, the bone mineral densities(BMD) were detected, the results were compared with blank control group. After establishing the animal model of osteoporotic fracture (OPF), we established the intramedullary nail fixed model. The rats which received fracture intramedullary nails fixation were divided into two groups at random:alendronat group and placebo group. Alendronat group was given suspensions of alendronat at does of5mg/kg/w for4weeks, while placebo group was given physiological saline at does of10mL/kg/w for4weeks. The X-ray films were performed at the end of2and4weeks after OPF for evaluating the bony callus and the fracture line. At the same time, the histopathological examinations and HE stains were performed, the BMD of new bony callus from the place of femur fracture were examined with the Figure Analysis System. All data were analyzed by SPSS17.0software.Results:1. By comparison at the end of two weeks, the fracture lines in X-ray films in both alendronat group and placebo group were clearly visible, there were not apparent new bony callus, the two groups had no difference (P>0.05). After four weeks, the fracture lines in alendronat group almost disappeared, while those in placebo group were indistinct. Alendronat group has larger new bony callus than placebo group (P<0.05). The comparison of Gray Scale Score of fracture site:At four weeks, alendronat group’s Gray Scale Score at fracture site is obviously higher than that of placebo group (P<0.05)2. Histology examinations:At two weeks, the Trabecular Bone Density Values in the two groups presented no obvious distinction (P>0.05). However, the Trabecular Bone Density Values in alendronat group were obviously higher than that of placebo group in four weeks (P<0.05)Conclusion:Alendronate can obviously increase the new bony callus during the osteoporotic fracture healing of rat, it can also increase the bone density of callus, the result probably means that alendronate could promote the healing of rat’s osteoporotic fracture. |
