Objective:To research the effectiveness of retinoic acid induction of osteoporotic rats treated by alendronate,Drynaria Rhizome Extracts and their combination by the biomechanics and fracture healing;And the development of Drynaria fortunei cataplasm.Methods:(1)Experimental study on the effects of Drynaria Rhizome Extracts on osteoporosis fracture healing and bone biomechanics in rats.35 female SD rats were treated with retinoic acid 80mg.kg-1.d-1 by gastrogavage for 15 days to induce osteoporosis,after 5 rats were approved osteoporosis,the rest 30 rats whose right femur were cut by rongeur and fixed with φ1.0mm k-wire intramedullary.After surgery which were randomly divided into 2 groups 15 each.Control group gastrogavage with NS 8ml.kg-1.d-1;Drynaria Rhizome Extracts group gastrogavage with Drynaria Rhizome Extracts 90mg.kg-1.d-1.Five rats in each groups were killed at the second,forth and sixth week after surgery respectively to carry out for biomechanic tests,histopathologic examination and callus volume calculation.(2)Effects of alenophosphate and Drynaria Rhizome Extracts on fracture healing and bone biomechanics of osteoporosis rats.65 female SD rats were treated with retinoic acid 80mg/kg·d by gastrogavage for 15 days to induce osteoporosis,after 5 rats were approved osteoporosis,the rest 60 rats whose right femur were cut by rongeur and fixed with φ1.0mm k-wire intramedullary.After surgery which were randomly divided into 4 groups 15 each.Control group gastrogavage with NS 8ml/kg.w;Alendronate group gastrogavage with alendronate 40mg/kg.w;Drynaria Rhizome Extracts group gastrogavage with Drynaria Rhizome Extracts 90mg/kg.d;Combination group gastrogavage with alendronate 40mg/kg.w and Drynaria Rhizome Extracts 90mg/kg.d.At 2 weeks,4weeks,6weeks after surgery 5 rats of each group were sacrificed;respectively for biomechanic tests,adiological examination,histopathologic examination and bony callus volume calculation.(3)The development of Drynaria Rhizome Extracts cataplasm.Extraction of Drynaria Rhizome Extracts,Host material of cataplasm were gum Arabic,tragacanth gum,glycerin,pressure sensitive adhesive,zinc oxide by 2.0:2.0:4.0:0.6:0.1 the optimal proportion,and join the transdermal promoter:azone and propylene glycol as promoting agent,and the dosage was 4%.Preparation process according to the following steps:(1)the alcohol extraction:take rhizoma drynariae herbs smash into coarse particle(10 eyes),add 95%ethanol,extracting 3 times,the first time for 2 hours,the second times for 1 hour,the third times for 40 minutes,hot filtering,merging alcohol liquid,recycle ethanol concentration into concrete(relative density 1.23)and set aside(2)the preparation of substrate:gum Arabic,west astragalus adhesive,pressure sensitive adhesive,zinc oxide by add amount water swelling,heat to dissolve,in 50℃water bath,stirring constantly,join the zinc oxide.as well as the mixture of azone,propylene glycol,glycerin(azone.propylene glycol,glycerin in mortarr)mix for a quick.(3)the cream and coating:take herb extraction of extract and matrix transdermal agent in 50℃water bath,stirring mixing 20-40 minutes,the coating on the non-woven fabric,lining,cut into 8*10 size.Results:(1)Biomechanical results:(a)Elastic ratio:2 weeks postoperatively,alendronate,Drynaria Rhizome Extracts and their combination are better than Drynaria Rhizome Extractsalone,P=0.022;The alendronate group is better than control group,P<0.05.(b)Maximum load:alendronate,Drynaria Rhizome Extractsand their combination can improve biomechanical properties of the maximum load at 4 weeks after surgery,P<0.05.(2)Results of callus:2 weeks postoperatively,the callus was smaller in the Drynaria Rhizome Extracts group compared with the control group,P=0.007;the same results in the alendronate group P=0.004 and the combination drug group P=0.027.4 weeks postoperatively,the callus was bigger in the alendronate group compared with the control group,P= 0.036;the same results in the Drynaria Rhizome Extracts group P<0.001 and the combination drug group P=0.011.The callus was smaller in the Drynaria Rhizome Extracts group compared with control group,P=0.005;the same result in the alendronate group P<0.001 and the combination drug group,P=0.015.There are no different at 6 weeks after surgery.(3)X-ray result:At 2 weeks after fixation,x-ray showed a transparent zone in the osteotomy gap indicated no ossified calluses formed at that time.Compared to the control group,a wider translucent zone was evident between the broken ends of the right femurs from the experimental groups.At 4 weeks,radiographs revealed external callus production in the combination group,and the fracture line had become blurred.However,the osteotomy gap was found in the control group,the remaining gap was still visible in the control group at 6 weeks post-surgery.The combination group showed a continuous reduction in size in radiographs with no bridging calluses evident.(4)Histological results:In the control group and the alendronate group,the number of osteoclasts and osteoclasts were significantly lower than in other groups.Drynaria Rhizome Extracts group and combination group the original surface of osteoblasts and bone trabecular bone in lacunae within the number of osteoclast significantly more than the other groups,cartilaginous callus replace fibre bone,cartilage callus to osseous callus fast at the same time.(5)Cataplasm results:Mixed modulation matrix and promoting agent,per gram of plaster containing 9 mg rhizoma drynariae and plaster containing 27 mg per gram rhizoma drynariae,we got two dosage forms cream.ConclusionRhizoma drynariae and alendronate can effectively increase of retinoic acid induction of osteoporotic rats femur biomechanical performance,including the maximum load,compression stiffness and elastic ratio.Drynaria Rhizome Extracts can promote fractures healing.Alendronate and rhizoma drynariae combined treatment has an additional effect of osteoporotic rats fracture healing and callus formation.The development of Chinese medicine cataplasm is feasible and provides a new form of Chinese medicine.SignificanceThe results showed that Drynaria Rhizome Extracts was beneficial to the bone healing of osteoporotic fractures,and enhancement of bone biomechanical strength.The combined use has an additional effect of osteoporotic rats fracture healing and callus formation.We had developed the world’s first Drynaria Rhizome Extracts cataplasm.In ancient China there are already "transdermal drug delivery technology" is commonly known as "snake oil" the principle of which is the same,the disadvantages of "snake oil" is that the patches are not up to the demands of the transdermal drug delivery technology in today’s world:skin permeability,poor paste,paste repeatedly times less,no water,especially serious is transdermal drug delivery of drug utilization is low and allergic to human body.However,the new,natural and synthetic macromolecular stromal cataplasm,which avoids these shortcomings,will become a widely used carrier for the future pharmaceutical industry.From cataplasm such as western medicine flurbiprofen cataplasm great success in recent years,according to forecast of related institutions,in the next 20 to 30 years,there are about more than 30%of the drugs will change to transdermal drug delivery preparation,is bound to bring about a disease within a new upsurge of external treatment.But our country has rich and curative effect of patients apply agent resources and long history of traditional Chinese medicine external treatment,we believe that increasing as the substrate material varieties,cataplasm research of traditional Chinese medicine increasingly in-depth,it will be developed potential huge market.And as the population ages,more and more become the focus of attention of osteoporotic fracture,rhizoma drynariae can treat osteoporosis fracture,can promote the fracture healing,improve bone biomechanical properties,to treat vitiligo pharmacopoeia mentioned at the same time,external use is safe,if made the cataplasm paste preparation and follow-up study on the economic and social benefits will be limited.We have prepared the first of the world’s first pieces of Drynaria Rhizome Extracts cataplasm,and further research is continuing.
