Therapeutical Effects And Mechanisms Of Fuzhenghuayu Capsule In Urine Schistosomiasis Liver Fibrosis

Objective: To investigate the therapeutic efficacy and the mechanism ofFuzhenghuayu Capsule in murine schistosomiasis liver fibrosis.Methods: The models of liver fibrosis induced by S.japonicurn were establishedby exposure of mice percutaneously through the tail to cercariae of S.japonicum. Thekunming mice were randomly divided into normal control group(A), model controlgroup(B), Praziquantel-treated group(C), positive control by INF-γ-treated group(D), lowdose Fuzhenghuayu Capsule-treated group(E), middle dose Fuzhenghuayu Capsule-treatedgroup(F) and high dose Fuzhenghuayu Capsule-treated group(G). On6weeks after theinfection were orally introduced with praziquantel. Then treatment with FuzhenghuayuCapsule. Fourteen weeks after being infected, the animals were killed, serum transaminasewere detected by automatic biochemical analyzer. The serum concentration of Hyaluronicacid (HA), Laminin (LN), Procolagen Type III (PC-III) and Collagen Type IV(C-IV) wereassayed with Radioimmunoassay. The content of hydroxyproline(Hyp) were examined byChemical colorimetry. Histological sections of liver were evaluated by HE and Massonstains, Immunohistochemical techniques were used to detect the expressions of PDGF-BB,PDGFR-β and type I, III collagen.Results:1. The murine schistosomiasis liver fibrosis in mice emerged at the end of the6weeks.2. At14weeks, compared with the normal control group, the level of serum ALT, AST,HA, LN, PC-III, C-IV in the model group were significantly increased (P＜0.01), the areas ofegg granuloma and collagen were also increased obviously (P＜0.05), the level of Hyp andthe expression of PDGF-BB, PDGFR-β and type I, III collagen in murine liver tissue wereincreased significantly (P＜0.01). 3. At14weeks, compared with the model group, the level of serum ALT,AST, HA,LN, PC-III, C-IV in Fuzhenghuayu Capsule-treated groups and INF-γ-treated group weredecreased obviously (P＜0.05), the level of Hyp and the expression of PDGF-BB,PDGFR-β and type I, III collagen in murine liver tissue were decreased significantly (P＜0.05), the areas of egg granuloma and collagen were also decreased significantly (P＜0.05).Conclusions:1. Exposure of mice percutaneously through the tail to cercariae of S.japonicum for6weeks can set up the early hepatic fibrosis mice model.2. The Fuzhenghuayu Capsule have effective treatment on murine schistosomiasis liverfibrosis.3. Fuzhenghuayu Capsule have obviously inhibitory effect in the liver fibrosis inducedby Schistosoma japonicum infection. The mechanisms maybe related to improve liverfunction, reduced the expression of PDGF-BB and PDGFR-β.
