Objective:1.Discuss the relationship between intestinal flora and ethanol metabolism,then screen out strains with high metabolic capacity.2.Analyze the effect and mechanism of different probiotics on acute alcohol intoxication in mice.3.Explore the difference of relieve effect on acute alcohol intoxication mice treated with the same species probiotics in separate sources.4.Study the influence and mechanism of combined probiotics on acute alcohol intoxication in mice.Methods:1.Determin the human excrement and feces of ADH,ALDH activity by colorimetric.2.Screened candidate strains of probiotics by bacteria isolation and identification technology.3.The mice model of acute alcohol intoxication was established by the oral administration of ethanol.4.Alcohol intoxication time and intestinal tissue was observed and recorded.5.Detect and record the changes of blood glucose in mice by blood glucose meter and test paper.6.H&E staining was used to observe the intestinal and liver injury.7.The BAC,ADH,ALDH,acetaldehyde,LPS and ALT was measured by ELISA.8.The SOD,CAT and MDA were determined by colorimetric.Results:The relationship between the intestinal flora and ethanol metabolism ability,the screening for probiotic candidate strains.Fecal ADH activity of good drinkers was significantly higher than wimpy drinkers(p<0.01),ALDH was not be detected.Fecal whole bacteria ADH activity of good drinkers was significantly higher than wimpy drinkers(p<0.01),ALDH was not be detected.The enzyme activity in different strains was different.ADH in Lactobacillus plantarum of the poor drinker group(Low enzyme Lactobacillus plantarum,LLP group)activity was lower than the capacity in good drinkers(High enzyme Lactobacillus plantarum,HLP group).The effect and mechanism of intoxication time of Bifidobacterium group(B group),yeast group(Y group)and HLP group decreased significantly(p<0.01,p<0.05,p<0.05).The plasma BAC levels in all groups were lower than in A group at 180min.BAC was significantly lower in the B group,HLP group and LLP group at 270min(p<0.05).In A group,the blood glucose of mice decreased after transient elevation.The blood glucose of B group,Y group and the HLP group were relatively stable,and tended to be normal graually.The change of blood glucose in LLP group was higher,and changed rapidly from high to low.1.2 The content of ADH in the alcohol group did not change.There was an increasing trend in B group and HLP group.The activity of ALDH in the intestinal tract was low in A group.ALDH activity of B group and Y group in intestinal increased significantly(p<0.05,p<0.01).ALDH activity in intestinal tissue in A group decreased significantly(p<0.05).B group increased significantly(p<0.01)and Y group and HLP group increased.Increasing the content of acetaldehyde of intestinal in A group,B group,Y group and HLP group,the content of intestinal acetaldehyde decreased.Reducing the activity of SOD and CAT in the intestinal tissue in A group,content of MDA was increased.Activity of SOD in Y group increased significantly(p<0.01),Y group and HLP group CAT activity increased significantly(p<0.01,p<0.05)and the content of LLP group MDA decreased significantly(p<0.05).Intestinal LPS was elevated significantly in A group(p<0.05).B group and HLP group the LPS decreased.Tissue observation,A group were suffered severe intestinal damage,intestinal hyperemia,and intestinal wall was transparent.B group,Y group and HLP group,intestinal integrity were highest in comparison with LLP group.1.3 The content of ADH in the liver of A group increased.In B group,Y group and HLP group had an increasing trend.A group of mice in the liver of ALDH activity decreased.B group,Y group and HLP group increased significantly(p<0.01).Liver SOD and CAT activity in A group decreased significantly(p<0.05,p<0.01),the content of MDA increased.SOD and CAT activity in liver increased,the CAT of HLP group increased significantly(p<0.05),and the content of MDA decreased.ALT of A group increased significantly(p<0.01).In the B group,Y group and HLP group had a decreasing trend.1.4 SOD(p<0.01)and CAT activity of A group decreased,and the content of MDA increased in brain.The activity of SOD and CAT increased,and the content of MDA decreased.2.The effect and mechanism of combined probiotics on acute alcohol intoxication in mice2.1 Compared with the A group,alcohol intoxication time of Yeast combine with High enzyme Lactobacillus plantarum group(Y&HLP group)decreased significantly(p<0.05).At 180 min,the plasma BAC of each group decreased.At 270 min Y&HLP group decreased significantly(p<0.05).2.2 ADH content and the activity of ALDH of intestinal in Y&HLP group increased significantly(p<0.01).In A group,the content of acetaldehyde increased.Y&HLP group decreased significantly(p<0.05).The SOD and CAT activity of intestinal tissue in A group decreased,the content of MDA increased.Y&HLP group SOD activity increased significantly(p<0.05),the content of MDA decreased.The content of intestinal LPS in A group increased.HLP group and Y&HLP group,content of intestinal decreased significantly(p<0.05).HE observation,the villi in A group become short with serious damage.The shape was complete in Y&HLP group and the arrangement was neat,the nap damage was smaller.2.3 Content of liver ADH in Y&HLP group increased,ALDH activity increased significantly(p<0.05).SOD activity in the liver of A group decreased,CAT increased,the content of MDA increased.The SOD and CAT activity in the liver of each group increased,the content of MDA decreased.ALT in alcohol group increased significantly(p<0.01).Y&HLP group ALT decreased.HE observation,A group of mice liver cells arranged in disorder,swelling,unclear boundary.HLP group and Y&HLP group have a better recovery.2.4 Brain SOD and CAT activity in A group was decreased,the content of MDA increased.The SOD activity in Y&HLP group increased significantly(p<0.01),the content of MDA decreased significantly(p<0.05).2.5 The above research in Y&HLP group indicators were better than HLP group,but there was no significant difference.Conclusion:1.The ability to drink is related to the intestinal ethanol metabolism enzyme.The probiotics from different environmental have different level of ethanol metabolic enzyme.The same species strain from different sources has different level of ethanol metabolic enzyme.2.Bifidobacterium pseudocatenulatum,Saccharomyces cerevisiae and Lactobacillus plantarum with high enzyme have significant remission of acute alcoholism.3.The ability to alcohol metabolism from Lactobacillus plantarum with high enzyme of the strong drinking ability is higher than that in the Lactobacillus plantarum with low enzyme.4.The alcohol metabolism effect of combined strains is higher than that of single strain. |
