With improved quality of life, people’s idea to the wine has changed from the high-proof,stimulative and hard drinks to the low-proof,nutritional and hygienical wine. Yellow wine is a kind of low-grain and low-alcohol brewage,it is of the Chinese national characteristics,as one of the three most ancient wine products in the world,it enjoys the honor of "state wine". In addition,Yellow wine is known by its nutrition and health value. Zhuyeqing liquor is a great Chinese wine,and the mellow taste and the health effects had been affirmed by people since ancient times. In1998,zhuyeqing liquor was designated as the only health wine.As representation of health wine,the antioxidant properties of yellow wine and zhuyeqing liquor were measured by four indicators:activity in anti-linoleic acid peroxidation,DPPH free radical scavenging,superoxide anion radical scavenging and reducing power. The experiments were divided into the following groups:low-dose group,medium-dose group,high-dose group,blank control group and positive control group. The content of phenolic compounds in yellow wine and zhuyeqing liquor was determined by Folin-phenol method,in order to establish the antioxidant evaluation system.Then,volatile and concentrated components were isolated from yellow wine and zhuyeqing liquor through the rotary evaporator. The antioxidant properties of volatile and concentrated components were measured by established antioxidant evaluation system,in order to determine the part with a high antioxidant activity.Then,the above determinate part was selected as research material in this work,which was extracted to acquire different polar fractions of yellow wine and zhuyeqing liquor. This work confirmed the relationship between free radical scavenging activity and the total phenolic content of different polar fractions of yellow wine and zhuyeqing liquor.Finally,based on the positive correlation between free radical scavenging activity and the total phenolic content,10kinds of familiar phenolic substances were selected as standard samples in this work. The experiments were measured to determine specific phenolic compounds by HPLC analysis,so as to provide scientific foundation for the antioxidant properties of yellow wine and zhuyeqing liquor.The results showed that:(1)That activity in anti-linoleic acid peroxidation, superoxide anion radical scavenging,DPPH free radical scavenging and reducing power is a better antioxidant evaluation system,which can evaluate the antioxidant properties of yellow wine and zhuyeqing liquor objectively;(2)Yellow wine and zhuyeqing liquor had strong activity in anti-linoleic acid peroxidation,DPPH free radical scavenging,superoxide anion radical scavenging and reducing power. And the antioxidant properties increased with increasing concentration of samples. There were165.1μg/mL and85.2μg/mL phenolic compounds in yellow wine and zhuyeqing liquor,which explained the higher antioxidant activity of yellow wine than zhuyeqing liquor to a certain degree.(3)Volatile component did not contribute to yellow wine’s antioxidant activity,which focused on concentrated component; volatile and concentrated components had contributed to the Zhuyeqing liquor’s antioxidant activity,but concentrated component did more. Phenolic compounds played a very important role in the antioxidant activity of yellow wine and zhuyeqing liquor.(4)Different polar fractions had a varying degree of free radical scavenging activities,and the antioxidant activity increased with the concentration of samples. To yellow wine,ethyl acetate extract and acetone extract showed the highest scavenging activity; ethyl acetate extract,acetone extract and water-soluble were more active components to zhuyeqing liquor. Meanwhile,phenol results showed that the relationship between free radical scavenging activity and the total phenolic content of different polar fractions was positively correlated,and the correlation coefficients of yellow wine were0.9908ã€0.9866and0.9926;the correlation coefficients of zhuyeqing liquor were0.9892ã€0.9985and0.9912,which explained yellow wine and zhuyeqing liquor played antioxidant activity by phenolic compounds.(5)The antioxidant substances of yellow wine were (+)-catechin and syringic acid,whose contents were35.11%and29.97%respectively; The antioxidant substance of zhuyeqing liquor were syringic acid and (+)-catechin,whose content were36.81%and29.42%respectively.(+)-Catechin and syringic acid were the antioxidant substances of yellow wine and zhuyeqing liquor,which explained the antioxidant properties of yellow wine and zhuyeqing liquor. |
