The honewort is a traditional Chinese medicinal material as well as of staple and popular crude drugs in China, which belongs to Bupleurum L. of Umnelliferae. Honewort also includs the Chinese Pharmacopoeia of the original plant in the genuine medicine Bupleurum. The roots or the whole grass of honewort are used as medicine, and their medicinal value is extremely high. The honewort is widely used in clinic, because it has the efficacy of reconciliation fever, anti-inflammatory, anti-virus,t0protect liver cells, and to improve immunity. In recent years, the "return to nature" has swept the globe, health and beauty features continue to be developed, a substantial demand of market for commodities herbs is increased. However, due to years of excessive and indiscriminate digging and disorderly use of honewort, wild resources have been seriously damaged, so artificial planting of honewort has become a major source of supply. Now the issue of hybrid varieties, low germination rate and the lack of local varieties suitable for the climate conditions seriously hamper the development of honewort medicine industry.Shanxi province is one of the main wild honewort resources in distribution and home-grown honewort production areas. Plant tissue culture technology is an effective method for the rapid propagation of the good breeding of medicinal plants and elite germplasm. Taking in sixteen kinds cultivated, wild plants and test-tube plants from thirteen production areas in Shanxi province as the research objects, by observing the similarities and differences between various types of honewort test-tube plants growth, relatively exogenous plant hormones and different medium for rapid propagation of the role of test-tube plants, analysis of the dynamic saikosaponin contents in test-tube plants and seed plants of honewort, determination of saikosaponin for wild and cultivated. Provide a scientific basis for the honewort large-scale rapid propagation and selection of excellent germplasm resources.The growth potential of the observations and statistical results showed that same or similar traits with the type of wild or cultivated plants to the subculture of honewort test-tube plants, there were differences between the different types, genetic stability was maintained most of the test-tube plants.The results showed that of the role of exogenous plant hormones in test-tube plants growth and development:6-BA, KT, NAA, IBA and DSC in the certain range of concentrations on the growth of the test-tube plants were positively correlated, and above a certain value, expressed as inhibition of test-tube plants growth.1.5mg/L6-BA and0.8mg/L KT of role in promoting growth in the test-tube plants of the Lingchuanliuquan. NAA, IBA and DSC roots of the induction of the test-tube plants,1/2MS+0.1mg/L NAA+0.5mg/L IBA+1.5mg/L DSC, production since of Ruicheng the test-tube plants’root and shoot growth in good condition, with excellent agronomic characters.We found that through the rapid propagation medium optimization and screening:There are both positive and negative effects of Ag~+for the role of test-tube plants growth,1.4mg/L played a good role in promoting the growth for test-tube plants of honewort from the Lingchuanliuquan. With subculture and rooting of test-tube plants, in medium supplemented with3%and5%sucrose was appropriate, respectively. Compare with solid medium, liquid culture in test-tube plants the stem was straight, coarse and more buds.Test-tube plants and seed plants saponin content of dynamic analysis results show that, test-tube plants of saponin content was mainly between3~6months after rooting. The annual seed plant in field, in the gradual accumulation of saponins from the April of this year until the frost solar terms content decreased. Transplanting test-tube plants in field exhibited excellent traits, until solar terms the beginning of winter were still synthetic saponin. With saikosaponin a, saikosaponin d and total saponins and other factors integrated examine, comprehensive advantages in the16kinds of wild and cultivated honewort are from Shanxi Yangqu, Jishan, Wanrong Lingchuanliuquan, Zuoquan and Xiaxian.
