Rationales:Bupleurum a common Chinese herbal medicine,has good therapeutic effects in soothing the liver and relieving depression,relieving the surface and fever.It is used widely and its inherent quality affects the efficacy of the prescription.In recent years,the resources of wild Bupleurum have become decreasing.It is advisable to find a medicinal material that can replace wild Bupleurum.Artificially culture of Bupleurum can remedy for the market gap which cause by significant reduction of Bupleurum.However,the content of effective components was low in artificial Bupleurum.Due to the influence of production place and cultivation technology,the sales quality of Bupleurum has changed a lot.Market is imbalance.This suggests that we should pay attention to the difference of internal quality of Bupleurum.It is caused by the origin and variety of Bupleurum.Therefore,by comparing the quality of Bupleurum cultivated in Shanxi with other varieties,different cultivation methods and different producing areas,it is necessary to establish a high-quality producing area of Bupleurum to improve the internal quality of Bupleurum products and maintain the safety and effectiveness of clinical medication.On the other hand,the Chinese Pharmacopoeia clearly stipulates that the medicinal part of Bupleurum is root.However,because of the rising price of bupleurum,,the non medicinal parts in the market are mixed with Bupleurum herbs and pieces to varying degrees.Although the chemical constituents of different parts of Bupleurum are similar,their contents are quite different.Excessive addition of non medicinal ingredients will inevitably affect the quality of Bupleurum.The effectiveness is reduced.So,it is of great significance for the quality control of Bupleurum to find out the different components in different parts of Bupleurum and establish the adulteration limit of non medicinal parts.The planting area of Bupleurum in Shanxi Province ranks the second in China,and Bupleurum chinense is the main variety.To control the quality of Bupleurum in Shanxi Province is of great significance to maintain the quality of the whole Bupleurum market.Objective:Establish a fingerprint that can simultaneously reflect the main chemical components of Bupleurum saponins,flavonoids,polyacetylenes,etc.,and find out by comparing the maps of Bupleurum chinensis grown in Shanxi with other different varieties,planting methods,and different producing areas.Different components,trying to clarify the characteristic components of Bupleurum spp.produced in Shanxi;and to compare the content of saponin components of Bupleurum spp.in different origins,varieties,planting methods and different parts of Bupleurum,providing a comprehensive evaluation of the quality of Bupleurum produced in Shanxi in accordance with.Methods:1.Investigate the resources of cultivated and wild Bupleurum in Shanxi Province,and collect samples of Bupleurum planted with reeds in various cities in Shanxi Province.For comparison,other varieties of Bupleurum,wild Bupleurum,and others were also collected.The province produces Bupleurum.2.UHPLC-Q-TOF-MS technology is used to identify the medicinal roots and non-medicinal basal stems of Bupleurum spp.The chemical composition is judged according to the reported literature mass spectrometry data and the cleavage law,and the chemical composition is determined.Compare the peak areas of the two to clarify the difference components.3.The methanol extract of Bupleurum Bupleurum was detected by HPLC technology.The detection time was 72 minutes,the detection wavelength was 254 nm,and the fluidity was 0.1% formic acid water(A)-0.1% formic acid acetonitrile(B),and gradient elution was performed,Constructed a fingerprint that can simultaneously determine the chemical components of Bupleurum saponins,flavonoids,polyacetylenes,etc.;and compare and analyze Bupleurum from different varieties,different planting methods,different producing areas and different parts through similarity analysis and multivariate statistical analysis.,Find out the mark peaks that can identify different varieties,different planting methods,different origins and different parts of Bupleurum.4.Using HPLC technology to determine the saponins content of the collected Bupleurum,and compare the saponins content of Bupleurum saponii grown in Shanxi with other different varieties,different planting methods,and different origins,and it is clear that the saponin components are in The differences between different varieties,different planting methods,and different producing areas of Bupleurum,and the content of saponins in different parts of Bupleurum are compared,in order to find the range for judging whether the adulteration of non-medicinal parts meets the standard.Results:1.Thirty-seven Bupleurum medicinal samples of different origins,different cultivation methods,and different origins were collected from the planting base,one of which was from Hebei Province,and the remaining 36 were from 8 cities in Shanxi Province(17 from Yuncheng City and 17 from Jincheng City).2 copies,3copies in Linfen City,3 copies in Changzhi City,2 copies in Jinzhong City,4 copies in Luliang City,1 copy in Xinzhou City,5 copies in Datong City),a total of 5varieties(31 copies of North Bupleurum,2 copies of South Bupleurum)Bupleurum,1portion for Tibetan Bupleurum,2 portions for Black Bupleurum,1 portion for Huang Bupleurum),including both wild and planted cultivation methods(planting Bupleurum 30 portions,’Wild Bupleurum 7 portions).2.Using UHPLC-Q-TOF-MS technology to detect different parts of Bupleurum,67 compounds were identified in the roots of Bupleurum,and 48 compounds were identified in the basal stems of Bupleurum,among which the different components were mainly saponin Comparing the peak areas of different parts of Bupleurum,it is found that the content of 5 components in the root of Bupleurum is significantly higher than that of the basal stem.All of these five compounds are saponins;there are7 components in the basal stem of Bupleurum.It is significantly higher than that in Bupleurum root.These components are flavonoids and chlorogenic acids.These components can be used as indicators to determine whether there is adulteration in non-medicinal parts.3.An HPLC detection method that can simultaneously reflect the fingerprints of the main chemical components of Bupleurum saponins,flavonoids,and polyacetylenes has been established,which can relatively comprehensively characterize the characteristics of small and medium-molecule compounds in Bupleurum.And by comparing the maps of different planting methods and different producing areas of Bupleurum in northwestern Shanxi,combining multivariate statistical analysis and ROC curve to predict the difference components,it may become the identification of different varieties,different planting methods,different producing areas of Bupleurum and different parts of Bupleurum Index component.4.The content of saponins in 37 batches of Bupleurum collected from the collection found that the content of saponins in Yuncheng,Datong and Changzhi grown in Shanxi is similar and higher than that in other areas,while the content of saponins in Luliang area is relatively low;Except for a batch of wild Bupleurum from Pangquangou Mountain,Jiaocheng County,Luliang City,Shanxi Province,the content of saponins in the planted and wild varieties of Bupleurum from other batches is not much different;by comparing different varieties of Bupleurum The content of saponins found that the saponins content of Bupleurum cylindrica and Bupleurum angustifolia are similar.The saponins content of Bupleurum black is more than that of other varieties of Bupleurum,while the content of saponins of Bupleurum pilaris is lower;in the underground part of Bupleurum In different parts of the saponins,the content of saponins in the roots was significantly higher than that in the basal stems.Conclusion:Through the comparative analysis of different varieties,different planting methods,different areas of Bupleurum and different parts of bupleurum,it was found that there were significant differences among different kinds of Bupleurum.On the one hand,we should comprehensively consider the evaluation index of the quality of bupleurum,in addition to the characteristic components and common components,the influence of non common components on the quality and efficacy of Bupleurum should also be considered;on the other hand,saponins,as an important active component in Bupleurum,its content is of great significance for the selection of high-quality Bupleurum and the quality evaluation of Bupleurum.
