ObjectiveTo evaluate the effects of moxibustion pretreatment at Baihui(GV20) and Shenshu(BL23) on the expression of NGF and BDNF in hippocampus CA1of the Alzheimer disease (AD) model rats. And to provide a experimental basis for the clinical treatment on this disease.Methods40male adult SD rats were randomly allocated and equally into normal, sham operation, model group and moxibustion pretreatment group which after40times moxibustion pretreatment at Baihui and Shenshu, bilateral hippocampus of moxibustion pretreatment and model rats were one-off injected with condensed5μl Aβ1-42for preparing the AD rats’model, while sham operation group were one-off injected with the equal physiological saline. Morris’water maze test was used to evaluate the rats’learning and memory abil ity. The expression of NGF and BDNF in the CA1region of the hippocampus was detected using immunohistochemical techniques.Results1. Morris water labyrinth test found model rats’ability of learning and memory had been damaged obviously. The model group compared with the normal group and the sham operation group is have significant difference (P<0.01); Compared with Normal and sham operation groups, the Moxibustion pretreatment and model groups in learning and memory ability during the Morris’water maze tests of place navigation and spatial probe.2. By immunohistochemical method, we can observe that the numbers of NGF and BDNF positive cells in hippocampus CA1of AD model rats compared with the normal group and the sham operation group were obviously reduced. While the Moxibustion pretreatment group is significantly superior to model group,but still lower than the normal group.Conclusion1. AD disease is caused by decline of kidney essence, the marrow of brain is absent and the spirit key can not be used of the old people. The treatment is based on the principle of tonic righting, particular emphasis on tonifying the kidney essence and filling up the marrow of brain, thereby achieving the purpose of the prevention and control of the AD. Baihui and Shenshu can be used as the main points of choice for the prevention and treatment of AD disease.2. Moxibustion pretreatment at the Baihui and Shenshu points can improve learning and memory ability of AD rats. The effect might be related with inceased NGF and BDNF espression in the hippocampal CA1region.
