| Objective:Set up aging rat medols.Give " BuShen YiJing" prescription Chinese medition Er Jingling (EJL),vitamin E and the nomal diet. Examine the capability of learning and memory. Then the activities of SOD, MDA, protein carbonyl levels of brain, liver, kidney and blood viscosity were measured. And the express of4-HNE protein in hippocampus, liver,kidney was determined.To observe the effects of BuShen YiJing prescription on indexes of carbonyl stress and to investigate the deferring aging effect.Try to explain the action mechanism of BuShen YiJing prescription on aging.Methods:44SPF wistar male rats, weighing200±20grams, were randomly divided into four groups as normal group, model group, EJL group and vitamin E group. At the same time gavage of EJL water decoction for EJL group, vitamin E for vitamin E group, physiological saline for normal group and model group, D-galactose was injected subcutaneously to prepare the aging rat models. Six weeks later, Morris water maze was used to examine the capability of learning and memory. Then the activities of SOD, MDA, protein carbonyl levels of brain, liver, kidney and blood viscosity were measured. Immunohistochemistry techniques were performed to determine the expression of4-HNE protein in hippocampus,liver, kidney. hematoxylin-eosin staining was used to observing the hippocampal micro-structure.Results:1. Morris water maze experimentIn the orientation course experiment, all rats escape latencies were shortened. Compared wi th the normal group, escape latency of the model group was significantly prolonged (P<0.05, P<0.01).In the Space exploration experiment, the target quadrant dwell time of model group and crossing platform number was lower (P<0.01), compared with the normal group,the target quadrant dwell time of EJL group was longer (P<0.01), crossing platform number of EJL group was larger(P<0.05) and20%outerring area percentage was smaller, compared with the model group.2. Blood viscosityCompared with the normal group, whole blood viscosity in low shear rate (blood1, blood5), secondary shear rate (blood50, blood100),high shear rate (blood200) of the model group increased significantly (P<0.05),plasma viscosity increased (P<0.01) and hematocrit increased significantly (P<0.05). whole blood viscosity, plasma viscosity, hematocrit of EJL group were reduced (P<0.05, P<0.01)3.Brain,liver,renal SOD activity,MDA content,protein carbonyl contentCompared with the normal group, SOD activity of model group was significantly reduced (P<0.01), MDA were increased (P<0.05, P<0.01), protein carbonyl levels were significantly elevated (P <0.01). Compared with the model group, SOD activity of EJL group was significantly elevated, MDA content decreased obviously, protein carbonyl levels were reduced (P<0.05, P<0.01).4. Immunohistochemical staining method for the detection of4-HNE protein expressionCompared with the normal group, IOD values of model group were significantly elevated(P<0.01). IOD of EJL group was significantly lower(P<0.01),compared with the model group.5. Morphology of hippocampus CA1region tissuePyramidal cells in hippocampus CA1region of Model group layers, with nuclear hyperchromatism, pyknosis, fracture. Layer of pyramidal cells which is staining clearly split neatly and distributes uniformly, densely,with pale-staining nuclear. cell morphology Vitamin E group and EJL group was significantly improved.Conclusion:Escape latency,20%outer ring area percentage was larger than that in the normal group. The target quadrant dwell time and crossing platform number was lower, compared with that in nomal group.So our rat models were successful.The experimental results showed that " BuShen YiJing " Method can significantly increased the SOD activity, decreased the MDA and protein carbonyl content, and improved blood viscosity state, the ability of learning and memory. It also decreased expression of4-HNE protein and improved hippocampal micro-structure. |
