Objective:To investigate the preventive and therapeutic effects of Rana Chensinensis skin extracts (RCSE) on three experimental gastric ulcers.Methods:â‘ Preparation of RCSE:The1000g of fresh Rana skin soaked at4°C in6times80%(volume fraction) methanol solution and replacement once methanol every24h. Combining the extracts and concentration. And pressure increase in dry methanol at37℃, dried for48h.â‘¡In an i.e. water immersion stressed mice test animals were randomly divided into model control(distilled water), cimetidine (0.2g·Kg-1)and three RCSE groups(0.22ã€0.45ã€0.90g·Kg-1).There were gavaged for15days and were measured the ulcer index(UI), GSH-Px activity levels of serum of liver. T-SOD activity levels of serum of liver and MDA levels of serum of liver.â‘¢Animals were randomly divided into model control, cimetidine (0.2g·Kg-1) and three RCSE groups(0.22ã€0.45ã€0.90g·Kg-1) in pylorus ligated rats test, They were orally administered with distilled water, cimetidine and RCSE for14days. There were gavaged for15days after create models of gastric ulcer and were measured the ulcer index(UI), GSH-Px activity levels of serum of liver, T-SOD activity levels of serum of liver and MDA levels of serum of liver.⑤In gastric ulcer in mice test, mice were pretreated by RCSE for7days, and then were administered with absolute ethanol orally to induce gastric mucosa lesions, the ulcer index reduced. The levels of malondialdehvd (MDA) reduced. The activity of T-SOD and the glutathione(GSH-Px) and the nitric oxide(NO) in the gastric mucosfl were raise. Chinese Forest Frog and L-NAME affect NO.Results:â‘ There are yield of8.15%RCSE and content of29.56%protein.In an i.e. water immersion stressed mice test, compared to the model control group, three doses of RCSE significantly decreased the ulcer index (UI) in water stressed mice (P<0.01), compared to0.22g·Kg-1and0.45g·Kg-1of RCSE groups,0.90g·Kg-1of RCSE significantly decreased the ulcer index (UI) in water stressed mice (P<0.01). Different doses of RCSE groups significantly increased the GSH-Px and T-SOD activity levels of serum of liver and significantly decreased and MDA levels of serum of liver(P<0.01). And compared to0.22g·Kg-1and0.45g·Kg-1of RCSE groups,0.90g·Kg-1of RCSE group significantly increased the GSH-Px and T-SOD activity levels of serum of liver and significantly decreased and MDA levels of serum of liver(P<0.01).â‘¢The0.60g·Kg-1RCSE group was changes on the UI in pylorus ligated rats test,(P<0.05). Three dose of RCSE group were significantly increased the GSH-Px and T-SOD activity levels of serum and gastric tissue of liver and significantly decreased and MDA levels of serum and gastric tissue of liver(P<0.01or P<0.5) in pylorus ligated rats test. Compared to the0.15g·Kg-1and0.30g·Kg-1RCSE groups, the0.60g·Kg-1RCSE group was significantly increased the GSH-Px activity levels of serum and gastric tissue of liver. Compared to the model control group, the0.60g·Kg-1RCSE group was significantly decreased the total acidity of gastric juice, total acid output of gastric juice and pepsin activity(P<0.01or P<0.5).â‘£Compared to the model control group, in an acetic acid induced rats test three dose of RCSE group were significantly increased the GSH-Px and T-SOD activity levels of serum of liver and significantly decreased and MDA levels of serum of liver(P<0.01or P<0.5). And there was significantly decreased the UI of0.30and0.60g·Kg-1RCSE group (P<0.05).0.60î—šKg-1RCSE group compared with the other groups was significantly increased the GSH-Px and T-SOD activity levels of serum of liver(P<0.01).⑤n gastric ulcer in mice test, mice gastric ulcer indices decreased after pretreated with RCSE, MDA contents of gastric mucosa were respectively all significantly lower than the model group. GSH-Px and NO contents were significantly increased in all the activities of T-SOD were significantly increased in all groups. L-NAME restrains NO, RCSE promote to reduce ulcer index.Conclusion:RCSE exhibits the preventive and therapeutic effects on the experimental gastric ulcers, and presents its anti-oxidation function. |
