Hypertrophic scar (HS) is severely affected people,s appearance and function, reducing the quality of life.So researchers have long tried to use a varietyof solution for the pathological scar, including surgery, drugs, radiation, pressure, and so on.But there is still not satisfactory curative effect.On the base of the previous studies of10-hydroxycamptothecin on human hypertrophic scar fibroblasts,we continue to explore the mechanism of HCPT on NF-κB expressionof human hypertrophic scar fibroblasts.Objective:In order to understand the mechanism of HCPT on treat scar, this studyobserved the effects of HCPT on NF-κB expression of cell transcription factor.Methods:HS obtained from patients who received plastic surgery, and then hypertrophicscar fibroblasts (HSFB) were cultured in vitro using tissue culture method. HSFB wasintervened by HCPT with various concentrations (125ã€250ã€500ng/ml) for24hours,then the expression of NF-κB p65ã€IκBα mRNA was measured with RTï¼PCR andNF-κB p65ã€IκBα protein was detected with western blot method.Results:(1) Compared with control groups0.104±0.032,0.974±0.039, the expression ofNF-κB p65〠IκBαmRNA in experiment groups is0.268±0.032ã€0.870±0.051,0.562±0.051ã€0.762±0.067,0.790±0.038ã€0.664±0.086with various concentrations(125ã€250ã€500ng/ml). There is significant difference between control groups andexperiment groups (P<0.05). These is indicated that hydroxycamptothecin couldsignificantly increase the expression of NF-κB p65mRNA and reduce theexpression of IκBαmRNA in human hypertrophic scar fibroblasts,especially when itsconcentration at500ng/mL.(2) Compared with control groups0.271±0.114,1.090±0.017,the expression ofNF-κB p65〠IκBαprotein in experiment groups is0.455±0.012ã€1.060±0.033,0.593±0.015ã€0.736±0.016,0.765±0.023ã€0.555±0.021with various concentrations (125ã€250ã€500ng/ml). There is significant difference between control groups andexperiment groups (P<0.05). These is indicated that hydroxycamptothecin couldsignificantly increase the expression of NF-κB p65protein and reduce the expressionof IκBαprotein in human hypertrophic scar fibroblasts, especially when itsconcentration at500ng/mL.Conclusion:Hydroxycamptothecin could reduce the expression of IκBαmRNA andprotein,which to enhanced the expression of NF-κB p65mRNA and protein, whichhas a role in the treatment of hypertrophic scars. |
