Background:Hypertrophic scar(HS)is a common disease in plastic and cosmetic surgery,and its pathogenesis is still unclear.Even though various clinical treatment options are available,none can achieve satisfactory results.Therefore,there is an urgent need to find a new strategy that can solve the patient’s suffering or even cure HS.Objectives:This study aimed to investigate the effects and mechanisms of emu oil(EO)on HS from both cellular and animal experiments.Methods:We extracted primary fibroblasts from human HS and identified the extracted primary cells;used EO to intervene in human hypertrophic scar fibroblasts(HFs);determined the appropriate concentration of EO by detecting the cell morphology,viability,short-term adhesion,and proliferation of HFs;examined the effect of EO on cell cycle and apoptosis of HFs using loss cytometry;Cell scratch assays were performed to investigate the effect of EO on the migratory ability of HFs cells,which was verified again using Transwell assays.After the successful establishment of the HS model,the rabbit ears were wet-dressed with 0.9%Nacl or EO and photographed after 4 weeks;the scar thickness of each group was measured,and the expression of pro-fibrotic factors and related collagen in each group was detected;histological staining of the samples was performed to assess the pathological damage and collagen deposition results,and the effect of EO on HS in rabbit ears was analyzed.Results:Based on the cell morphology and immunofluorescence results,the extracted cells were identified as HFs.0.02% EO inhibited the growth of HFs without affecting the cell morphology and cell adhesion,which was the appropriate concentration for administration;EO interfered with the cell cycle of HFs,but had no significant effect on apoptosis;EO significantly inhibited the migratory ability of HFs;EO attenuates HS formation in rabbit ears by decreasing the expression of pro-fibrotic factors and related collagen,reducing pathological damage and collagen deposition.Conclusion:We found that EO inhibits the proliferation of HFs,interferes with the cell cycle,weakens cell migration,and plays an essential role in combating HS fibrosis and remodeling the extracellular matrix.Therefore,the above findings suggesting that EO has excellent potential as a new strategy for treating HS.
