| The natural pigment is rich in resources, highly safe, non-toxic side effects and it has a variety of physiological activity such as antioxidant, scavenging free radicals, anti-tumor, anti-mutagenic. This article takes three northeast wild berries as raw material (Vaccinium Uliginosum, Padus virginiana, Prunus maackii). Comparative study of pigment aqueous extract and ethanol extract from three berries of the antioxidant activity in vitro. The antioxidant capacity after the purification of pigment from the three kinds of berries is compared with Vc, and after purification of pigment extracts from three kinds of berries effects on tumor cells, study on the inhibition of tumor cell. The results show as follows:According to the antioxidant activity in vitro tests on the pigment aqueous extract and ethanol extract from the three kinds of berries. The results show that pigment crude extracts from three kinds of berries can scavenge DPPH radicals, hydroxyl radicals, superoxide anion radicals and hydrogen peroxide radicals effectively, and it has a stronger total reducing capacity and total antioxidant capacity, but different extractive from same kinds of berries antioxidant capacity is diverse. Pigment ethanol extract from Vaccinium Uliginosum scavenge DPPH radicals capacity is the strongest, clearance rate is (93.50±3.28)%; Pigment aqueous extract from Padus virginiana scavenge hydroxyl radicals capacity is the strongest, clearance rate is (95.45±3.13)%; Pigment ethanol extract from Vaccinium Uliginosum scavenge superoxide anion radicals capacity is the strongest, clearance rate is (98.80±2.02)%; Pigment aqueous extract of Prunus maackii scavenge hydrogen peroxide radicals capacity is the strongest, clearance rate is (70.71±2.75)%; Pigment alcohol extract from Prunus maackii has the strongest total reducing capacity which is1.901; Pigment ethanol extract from Prunus maackii has the strongest total antioxidant capacity which is13.51units/mL.The pigment extracts from three berries after purfying take antioxidant activity in vitro test and compare with Vc. The results show that the antioxidant capacity after the purification of pigment extracts from three berries is stronger. The antioxidant capacity of them is less than Vc, besides the pigment extracts from Vaccinium Uliginosum scavenge DPPH radicals. Under the similar conditions of clearance rate, before pigment extracts purfying, the mass concentration can be several times or a few times after purfying. The antioxidant capacity is enhanced remarkably after the purification of pigment extracts from three berries than pigment crude extracts.The pigment extracts from three berries can inhibit on the growth of human colon cancer HT29cells, human hepatoma HepG2cells and human cervical cancer Hela cells. The inhibition rate increased significantly with the increased of the concentration of pigment and treatment time. After comparison, The pigment extract from Prunus maackii has the best inhibitory effect on HT29cells and Hela cells, the inhibition rates is (60.88±4.54)%and (64.37±4.08)%respectively. The pigment extract from Vaccinium Uliginosum has the best inhibitory effect on HepG2cells, the inhibition rates is (63.74±4.48)%.In three kinds of tumor cells the activity of GSH-PX, CAT, T-SOD, NOS and the content of NO decreased with the increasing concentration of pigment extrects from three kinds of berries. Moreover, it has a certain dose-effect relationship. The content of MDA increased with the increasing concentration of pigment extrects from three kinds of berries. Moreover, it is a linear relationship. This indicates that the pigment extracts from three kinds of berries can reduce the activity of antioxidant enzymes, increase the content of lipid peroxidation products in tumor cells, and reduce the activity of NOS and the content of NO, in order to inhibit on the growth of tumor cells. |
