Experimental Study Of Electro-acupuncture Combined With Astragalus Polysaccharide On Db/db Mouse’ Blood Glucose And Pancreatic Islet Beta Cell Function

Objective:Observation the effect of Electro-acupuncture combined with Astragalus Polysaccharide(APS) in treating type2diabetes and its effect of pancreatic islet beta-cell function, discussed its mechanism,to provide experimental evidence and new method for the clinical treatment of the disease.Methods:Taking db/db mice as experimental animal model,which were randomly divided into four groups:model group, Chinese medicine group, acupuncture group,acupuncture combined with medicine group, taking10db/m mice as normal control group,10rats in each group.The mice were observed daily diet, drinking water, urine, mental status and the activity, every7days to measure weight, regular testing blood glucose of tail vein. The traditional Chinese medicine group were fed with APS by gavage for one time daily,5times a week, according to700mg/kg body weight;The acupuncture group were pinpricked combined with electro-acupuncture, one time a day,5times a week,for11weeks;The acupuncture combined with medicine group were both fed with APS and pinpricked one time a day,for11weeks;the normal group and model group were not given any treatment. In order to ensure that all the experimental conditiongs are the same,the acupuncture group were given equal doses of distilled water by gavage,the Chinese medicine group were binded and fixed the same time. In experiment eleventh weekend,collecting specimens, fasting, free water based24h. Anesthetized by intraperitoneal injection to mice in each group, picked off all mice’eyeballs and collected their blood samples to detect the serum concentrations of blood glucose and insulin.Took the pancreatic tissue for observing their morphological changes under optical microscope.using immunohistochemical method for the determination of Bcl-2protein expression,which is closely related to islet cell apoptosis.Results:(1)Observing pancreatic tissue morphological similarities and differences of mice under optical microscope:Model group mice pancreas islet structure have changed:size and irregular shape, part of the cells were vacuolated degeneration, cell gap widened, the uneven distribution of individual cells, Each treatment group compared with model group, in the islets of Langerhans cell distribution is uniform, the cell size is consistent, the nucleolus is clear, complete, realm of islet node is clear. (2)serum glucose levels after treatment:Each treatment group’glucose level compared with the model group was significantly decreased,the difference had statistical significance (P<0.01);the acupuncture combined with medicine group was the lowest of all the treatment groups,the difference had statistical significance (P<0.01);the glucose level of acupuncture group is lower than the Chinese medicine group,and two groups have significant difference(P<0.01).(3)Insulin level after treatment: Insulin levels in each treatment group than the model group was significantly decreased, the difference had statistical significance (P<0.01);The insulin level of acupuncture group and acupuncture combined with medicine group was lower than the Chinese medicine group, the difference had statistical significance (P<0.01);acupuncture group and acupuncture combined with medicine group were no significant difference (P>0.05).(4)Islet beta-cell apoptosis related gene Bcl-2protein expression:Expression of Bcl-2in Each treatment group’compared with the model group was significantly increased,the difference had statistical significance (P<0.01); and the acupuncture combined with medicine group was the most significant in improving the expression of Bcl-2,the difference was significant (P<0.01);between acupuncture group and the Chinese medicine group, there was no significant difference(P>0.05).Conclusion:(1)Both acupuncture and medicine can protect normal morphology of islet tissue,improve function of islet cell;acupuncture combined with medicine is superior to the pure acupuncture or medicine.in protect ing morphology of islet tissue.(2)Acupuncture combined with medicine superior to the pure acupuncture or medicine therapy in reducing blood glucose levels,and acupuncture g is also superior to the Chinese medicine in reducing glucose levels.(3)Acupuncture combined with medicine is superior to the pure acupuncture, the Chinese medicine in decreasing the serum insulin level, so as to improve the insulin sensitivity of diabetic mice.(4)Bcl-2is inhibitory factor of cell apoptosis, acupuncture has the exact effect in treating diabetes,presumably,acupuncture may play a role by improving protein expression of inhibitory apoptosis factor Bcl-2and decreasing islet beta-cell apoptosis.And the combination of acupuncture and medicine can play a synergistic effect.
