Objective: To investigate the effect of intremedin 1-53 on IL-8 and IL-23expression in rats with acute lung injury induced by lipopolysaccharide.Methods: Forty -eight healthy Wistar rats were randomly divided into threegroups:normal controls group(C group), acute lung injury induced bylipopolysaccharide group(ALI group)and intermedin1-53-pretreated group(D group). .Group C, the intraperitoneal injection of normal saline 1ml, 1h after intravenousinjection of normal saline 1ml, ALI group of normal saline 1ml, 1h after intravenousinjection of LPS 5mg/kg (dissolved in 1ml saline), D group of the intraperitonealinjection IMD1-53 2nmol/kg (dissolved in 1ml saline), 1h after intravenous injectionof LPS 5mg/kg (dissolved in 1ml saline).At 2h, 4h two time points ,8 rats from eachgroup were killed and lung plasma obtained,Measured by ELISA serum levels ofIL-23; immunohistochemical method to measure tissue levels of IL-8, observed ineach group lung wet / dry weight ratio (W / D), to observe the morphological changesin lung tissue.Results: 1 IL-23 serum levels compared with C group, ALI group, serum IL-23were significantly increased, and increased with time, the difference was statisticallysignificant (P <0.05), D serum levels compared with IL-23 ALI group decreased, butstill higher than the C group, compared with the other two groups were statisticallysignificant (P <0.05). 2 lung tissue expression of IL-8 expression is weak in group C,ALI group was significantly enhanced, D ALI group compared with the weakexpression group, treated 4h group difference was statistically significant (P <0.05),2h group differences, but not significant may not fully 2h, intermediary factors play aprotective role, ALI group C was significantly (P <0.05). 3 lung W / DC group twotime points did not change significantly, the difference was not statistically significant,ALI group was significantly higher compared with C group at each time point was significantly (P <0.05), D group than in the ALI group , but was still higher than the Cgroup. Pathological changes in lung tissue 4: ALI group with varying degrees ofswelling of the lungs appear congestion, scattered severe bleeding; optical microscopyshows diffuse alveolar septal thickening, some alveolar collapse, interstitial diffusepulmonary hemorrhage, alveolar shows a large number of inflammatory cellinfiltration and exudate. Group D by IMD1-53 protection, lung congestion andswelling, inflammatory cell infiltration and alveolar exudate, alveolar septalthickening and the degree of alveolar collapse was significantly reduced comparedwith ALI.Conclusion: IMD1-53 has a protective role in acute lung injury in rats, theprotective effect may be through reduced IL-8, IL-23 expression of inflammatoryfactors such as implementation. |
