Objective: superoxide dismutase (SOD), glutathione peroxidase (GSH-Px),lactate dehydrogenase (LDH), total nitric oxide synthase (T-NOS) activities andmalondialdehyde (MDA) content were detected with the testicular tissue homogenate.To study effect of citric acid on antioxidant of testis in mice, assess of testicular cellmembrane lipid oxidation and cell biofilm permeability, and explore citric acid anti-oxidative stress mechanism.Methods:40health Kunming mice were randomly divided into control group(0.9%saline), low dose group (180mg/kg),middle dose group (270mg/kg) and highdose group (360mg/kg). All the mice were received an injection of0.2mLcorresponding reagent each week for3weeks. After the third injection of seventh day,all the mice were killed by cervical dislocation, superoxide dismutase (SOD),glutathione peroxidase (GSH-Px), lactate dehydrogenase (LDH), total nitric oxidesynthase (T-NOS) activities and malondialdehyde (MDA) content were detected withthe testicular tissue homogenate.Results:(1)The SOD activity: with citric acid dose increased, the SODactivitiy were showed decreasing trend. Compared with the control group, the SODactivity was significantly decreased in middle and high dose group (P<0.05), and theSOD activity was no significant change in low and high dose group (P>0.05);(2)The GSH-Px activity: Compared with the control group, the GSH-Px activitysignificantly decreased in low and middle and high dose group (P<0.05), the activityof GSH-Px content were no significantly change (P>0.05);(3)The T-NOS activity:Compared with the control group, different concentrations of citric acid treatment, theT-NOS activity was no significant change (P>0.05), with citric acid dose increased,the T-NOS activity was showed rising trend, increased by2.1%,5.5%and18.3% respectively, compared with the control group.(4)The MDA content: Comparedwith the control group, the MDA content was significantly decreased in the citric acidtreatment groups (P<0.05); with citric acid dose increased, the MDA content showedrising trend, compared with the low and middle dose group, the MDA content wassignificantly decreased in the high dose group (P<0.05).(5)The LDH activity:Compared with the control group, Compared with the control group, differentconcentrations of citric acid treatment, the LDH activity was no significant change(P>0.05), with citric acid dose increased, the LDH activity was showed decreasingtrend, decreased by8%,8.6%and10.4%respectively, compared with the controlgroup.Conclusion: The results suggested that a certain dose of citric acid can degradethe SOD, GSH-Px activity of testis in mice, may be consumed a large number of SODand GSH-Px in the oxygen free radicals, resulting in testis the SOD and GSH-Pxactivity significantly antioxidant mechanism, or the reduction of lipid peroxidation andthe stability in testicular cell membrane may be via NO pathway. But the study alsorevealed that, with citric acid dose increased, the LDH activity was showed decreasingtrend, testis membrane permeability may be increased, the stability of the cellmembrane may be affected.
