The human papillomavirus (HPV) is a wide spread virus that can cause manydiseases in keratinocytes of the skin or mucous membranes. High risk types of HPVare related to many cancers such as cervical cancer, esophageal cancer, laryngealcancer and so on. Incidence of esophageal cancer in China is very high. There aremore than400thousand new patients with esophageal cancer in the whole worldevery year, among them more than one half are in China. According to some studies,Esophageal cancer is the result of the cooperation of multiple factors, and HPV16and18are important pathogenic factor of esophageal cancer in China.The cervical cancer HPV vaccines are proteins, which can only induce specificantibody to prevent virus infection. They cannot do as therapeutic vaccines which canclear the infected cells and cancer cells. Till now, no prophylactic or therapeuticvaccines have been reported. In some study, HPV L1has been detected in esophagealcarcinoma and esophageal carcinoma cell line. So here HPV L1is used to developesophageal cancer.In this paper, the recombinant DNA vaccines and adenovirus vaccines whichcontain HPV L1gene are constructed. Then C57BL/6mouses are immunized with thevaccines. One week after the last immunization, humoral immunity is detected bypseudovirus neutralization assay and cellular immunity is detected by enzyme-linkedimmunospot assay. The results show that both HPV16L1and HPV18L1vaccinescan induce very strong humoral immunity, the neutralization titer of immune groupwhich is injected with109vp or1010vp recombinant adenovirus vaccines is above640, the neutralization titer of immune group which is injected with108vprecombinant adenovirus vaccines is above320. The HPV16L1vaccines can surelyinduce strong L1specific cellular immunity. Only weak cellular immunity reaction isdetected in the groups which are immunized with HPV18L1vaccines, and this isprobably caused by the incorrect T cell epitopes which are needed in enzyme-linkedimmunospot assay. HPV16L1139-147(AGVDNRECI) and L1355-364(YKNTNFKEYL)are H-2Dbrestricted T cell epitopes of HPV16L1.
