Oo-stat plays an important role in preventing rabbit coccidiosis. The best scheme of Oo-stat and prevention of coccidiosis in rabbit were studied in this paper.1 Study on the best scheme of oo-stat based on restraining Eimeria stiedai oocystsTo select effective anti-coccidian oo-stat, three compounding of oo-stat were chosen to restrain the sporulation of Eimeria stiedai oocysts. The unsporulated oocysts were cultivated in those drugs1:20.1:40.1:60.1:80、1:100.1:200.1:300.1:400 and 1:500 under about 20℃respectively.After 6h、12h and 24h, the drugs were centrifuged and separated, the oocysts were cultivated in K2Cr2O7 solution. The rate of sporulation was observed after being cultivated for 5-7d. The results of the experiment indicated that, the efficacy of the three compoundings under 20℃was good. Among the three compoundings, the rate of sporulation of the second and the third were both lower than that of the first(P< 0.05), all the rate of sporulation decreased notablely compared with the check, even 1:500 decreased about 30%. Besides, when time extended, the rate of sporulation decreased gradually. Moreover, the higher the concentration of drugs were, the better the controlling function of drugs were. The first compounding was chosen as the best scheme.2 The influential factor on the efficacy of oo-statTo study the influence of temperature and organic substance in rabbit manure on evaluation of efficacy of biocide, the unsporulated oocysts were cultivated in those drugs 1:20.1:40.1:80 under about 20℃and 4℃respectively. The rabbit manure including unsporulated oocysts were also cultivated in 1:20.1:40.1:80. The results of the experiment indicated the efficacy of 1:20.1:40.1:80 under 20℃was better than that of 4℃. The efficacy of biocide on rabbit manure was worse than only oocysts. It could draw a conclusion that, it causes obviously different results in temperature and organics substance influence test. 3 Experimental observation on the excystation of Eimeria stiedai in various concentration of oo-statTo test the efficacy of oo-stat on preventing liberating Eimeria stiedai sporozoites from oocysts.sporulated oocysts were cultivated in oo-stat 1:20、1:40、1:60 for 6h、12h and 24h respectively, after centrifuged and washed, a tissue homogenizer was used to breakdown the wall of oocysts, the suspension was centrifuged again and the pellet was cultivated in sporozoite releasing fluid which contained trypsin and deoxycholate in PBS, pH 7.4. At last the number of sporozoite and sporange were counted compared with the check. The results of the experiment indicated that, the efficacy of oo-stat on preventing liberating Eimeria stiedai sporozoites from oocysts is good, the rate of sporozoite liberation diminish more as compared with the check (P< 0.05). It could draw a conclusion that oo-stat 1:20and 1:40 had the better effect of prevention of excystation.4 The effects of oo-stat on pathogenicity of sporulated coccidia oocystsTo check the efficacy of oo-stat against rabbit coccidiosis, Rabbits (n=60) were randomly grouped in oo-cysts-treated (Ⅰ-Ⅵ),7%ammonia water-treated (Ⅶ-Ⅷ), infected and treated control(IX), uninfected and untreated control(X) group, groupⅠ-Ⅷwere artificially infected with rabbit coccidian oocyst (3×104) which had been cultivated in oo-stat 1:20、1:40、1:60 and 7% ammonia water for 12h、24h respectively. Then these rabbits were raised for 28d. The results of the experiment indicated that the average weight gains of the oo-stat 1:20、1:40 were174%、163%、160%、167% respectively, and the 1:60 were 72%、86% compared with control. This means the efficacy against rabbits coccidiosis of oo-stat 1:20 and 1:40 are more excellent than that of oo-stat 1:60. The average bodyweight in group 1:20 and 1:40 increased more as compared with that in group1:60 (P<0.05), as compared with that in groupVIwas significantly different(P<0. 01). It could draw a conclusion that the oo-cysts 1:20 and 1:40 had the effect of prevention of rabbit coccidiosis and improved the growth of rabbits.
