| Rose is the important plant of Rosa L.. It is not only the species for garden greening,but also the important material for natural aromatic plants and aromatic industray, whichhas high economic value. It has been widely used medicine industry, with certainmedicinal value.Agarose gel electrophoresis conditions were optimized in this study. Geneticrelationship of17Rosaceae plants is analyzed by ISSR. The results shows:①From57ISSR primers,13primers were selected to perform ISSR-PCR. Total479DNA bandswere obtained,of which221bands had polymorphism,the percentage of polymorphicbands (PPB)was0.43,4DNA fragments were amplified with each of the13primer.Total479DNA bands were obtained,of which13bands had polymorphism,andaverage3polymorphic bands per primer.②All the plants can be divided into fourgroups:the frist group was consisted of Rosa rugosa ‘Thunb in Pingyin’;the secondgroup was consisted of Rosa chinensis ’Viridiflora’,Rosa chinensis ’Pu Fu Hong’, Rosachinensis ’Comtesse dll cayla’,Rosa chinensis ’Si mian jing’,Rosa chinensis ’Huo yue zhongyue’,Rosa chinensis ’Yu Shi Zhuang’ and Rose hybrid bracteata ‘Mermaid’; the third groupwas consisted of Rosa hybrid tea ’Sonia’ and Rosa hybrid tea ’Yankee Dodle’; and thefourth group was consisted of Rosa gallica ’Versicolor’, Rosa chinensis ’Jin Ou FanLv’,Rosa chinensis ’Sophie’s perpetral’, Rosa hybrid climbingroses ‘Ten Shen Qi’, Rosachinensis ‘Yi Ji Fen’, Rosa chinensis ‘Jin Fen Lian’and Rosa chinensis ‘Da Fu Gui’.By reseraching on phenophase and agronomic characters, comprehesive study ondiseases and insect pests, fertilizer and water treatment and different types of pruning onthe fresh flower yield and investigation upon hardwood cutting, the adaptability of Rosarugosa ‘Thunb in Pingyin’ in ChongMing was evaluated and cultivation technique wasoptimized, which formed the special rose planting system in ChongMing and establishedthe foundation of industuialized production for rose. The results are as follows:(1) The growth and morphological character of Rosa rugosa ‘Thunb in Pingyin’ are the same withthose in place of origin, while the ability of waterlogging tolerance, high temperatureresistance and even the yield was obviously raised compared to original.(2) Byresearching on prune technology, appropriate prune between the time of defoliaton andgermination can help basal portion sprout new branch and increase yield in the next year.(3)As for the much annual precipitation which dense from April to September, the maindisease is black spot. Two kinds of pathogenic bacteria was rhizoctonia and alternaria spp,which were separated and identified from diseased rose.(4)The technique of hardwoodcutting covered by plastic film for Rosa rugosa ‘Thunb in Pingyin’ in ChongMing in earlyspring was primary mastered. The survival rate of branches shoots were high, the top rateof which can reach80%. It not only increased the propagation rate, but also decreased thecost of cutting propagation, which solved the problem to scale production of roseseedlings. |
