Hypsizygus marmoreus is one of the main well-known edible mushrooms that has been realized industrial production and could be realized commercial production in China. By the end of2008, the commercial production of Hypsizygus marmoreus had reached up to10585tons, which was close to Flammulina velutipes and Pleurotus eryngii. Also, Hypsizygus marmoreus increasingly took the fancy of the consumers, due to the superior nutritive, medical, and health care values.Spawn quality is the fundament to the development of edible industry. However, fecundity trial as the most effective classical method to evaluate spawn quality for a long time, greatly exposured shortcomings such as inevitable impacts by environmental and artificial factors. In recent years, isozyme, RAPD, RFLP, and other molecular biology techniques were applied in the evaluation of spawn quality, but there was no clear correlation between the results of these evaluation methods and the relavent charecters. Meanwhile, several studies emphasized that spawn quality could be evaluated from the biochemical viewpoint. Therefore, the present study is focused on evaluating anticipated indicates from the biomechanical viewpoint, so as to seek some brandnew ideas and approaches for the evaluation of spawn quality of Hypsizygus marmoreus.The resaults could be generally categorized under three headings as following.1. LBL evaluation method, based on liquid culture, visible spectra analysis and fecundity trial, was developed as an evaluation indicate. Also, liquid culture medium was determined (g/L):200.0Potato,20.0Lactose,2.0NH4NO3,0.5MgSO4·7H2O,1.5KH2PO4,0.06Bromothymol Blue (BTB), pH7.0. The results showed that the correlations on DR and yield per bottle, number of fruit bodies, and weight of single fruit body were statistically significant. Then, fitting equations on DR, chromogenic reaction and weighted relation coefficient were determined, respectively.2. LBL evaluation method, were explored to determine the correlations between results of LBL evaluation and main commercial characters of Hypsizygus marmoreus with the pileus diameter and thickness, stipe length and diameter. The results showed that, relevant and effective relationships between the number of fruiting bodies are formed, LBL evaluation effect with10.0-20.0mm and20.0-30.0mm range of pileus diameter,5.0-10.0mm and0.0-5.0mm range of pileus thickness,50.0-70.0mm and30.0-50.0mm range of stipe length, and0.0-5.0mm,5.0-10.0mm and10.0-15.0mm range of stipe length. Meanwhile, fruit bodies in these ranges were the major component of the total fruit bodies.3. The optimization of nutritional requirements of culture, based on liquid culture and visible spectra analysis, was studied for the effect of LBL evaluation on SIEF4104and SIEF4078. The optimal combination of liquid culture medium was determined (g/L):200.0Potato,20.0Lactose,2.0Malt extract,1.0MgSO4·7H2O,3.0KH2PO4,0.06Bromothymol Blue (BTB), pH7.0. Also, certain scope that carbon and nitrogen sources on effect of LBL evaluation was found. Moreover, compared with liquid culture media which added inorganic nitrogen sources, liquid culture media which added organic nitrogen sources generally showed lower DR values.LBL evaluation method used in this study would contribute to acceleration of strain improvement, conservation of breeding period, and development of evaluation method of spawn quality of Hypsizygus marmoreus. Meanwhile, LBL evaluation method was studied to seek the anticipated indicates of spawn quality of Hypsizygus marmoreus from the commercial characters viewpoint. Also, it provided a brandnew reference for the evaluation of other edible mushrooms. Moreover, LBL evaluation method could be applied to the construction of evaluation method and system for spawn quality of Hypsizygus marmoreus, development of resource database of superior or inferior spawns, and contribution of revealing the degenerate mechanism of spawn quality.Still, there were some weaknesses in this study, such as:LBL evaluation mechanism is unclear; the evaluation effect on vegetative growth was not significant; integrated evaluation on commercial characters was difficult to apply due to the relationships between different characters were complicated and vulnerable under different cultivate medium and cultivate environment, and so on. Therefore, relavent studies need to be further carried out.
