In recent years, bioactive peptides became the focus of research. This paper made full use of industrial production of almond meal produced the protein resources, reuse, enhance the added value of the products.With Almond protein as raw material in this paper, using enzyme hydrolysis to prepared the antihypertensive peptide with high activity, the main results are as follows:(1) By single factor experiment and Box-Behnken design response surface optimization experiment to determine the optimal enzyme solution conditions:pH value was12.48, temperature was56℃, substrate concentration was3.5%, enzyme concentration [E]/[S]%was1.3%, and in the best condition ACE inhibition was82.89%.(2) Using ultrasonic aided enzymatic hydrolysis, through the single factor experiment and orthogonal test to determine the optimal ultrasonic time for30min, ultrasonic power for60%, ultrasonic temperature was40℃, and in the best condition ACE inhibition was84.57%.(3) Through the ultrafiltration to determine molecular weights below3KDa of ACE inhibitory peptides its inhibition rate reach86.02%; Using cation exchange resins and anion exchange resins mixed bed to desalt, determined the best process parameters for desalting of ACE inhibitory peptides:pH value of hydrolyte4.5, hydrolyte flow rate10BV/h and between anion exchange resins and cation exchange resins3:2; the inhibition of ACE inhibitory peptides was48.23%after simulated gastrointestinal digestion, still had a high activity; with the powder of soybean peptides to the national standard GB/t22492-2008as a reference to determine the ACE inhibitory was accord with top level of national standard; Amino acid analysis showed that ACE inhibitory peptides of almonds contains17amino acids, including8necessary amino acids. Among them, the glutamic acid, aspartate contents highest,12.12%and7.50%respectively; by the experiment of SDS-PAGE electrophoresis, the molecular weight distribution was below2000Da.Preparation of antihypertensive peptides produced by these tests, had a high activity, but also provided a theoretical basis for the industrialized production of almonds antihypertensive peptides.
