Whereas almond mostly were used on oil manufacture and complex utilization deficiently,immense waste was worked, the new avenue and product append value were provided by biological active peptide vocation at the rapid speed extend maintain,a better economic benefit and social benefit were produced along out for complex utilization almond protein resource into that Sinkiang. This paper studied on the theory of alkali extraction and acid precipitation to extract the isolated protein from sweet almonds of Xinjiang, determined the optimum process conditions, and hydrolyzates isolated protein and prepares almond peptide from almonds, then desalinating to protease products and getting the peptide,the main results are as follows:The optimum conditions of first alkaline extraction of almond isolated protein was: the temperature of extraction 50℃, the rotary velocity 4000r/min,the ratio of solid and liquid 1:25, and the extraction time 70 min were obtained by both single factor experiment and orthogonal experiment. The extracting ratio is 78.3%. Extracted temperature and the ratio of solid and liquid is more important than the rotary velocity and the extraction time about extraction rates of almond isolated protein. The factors were studied that affected the preparation of almond peptides. The regression models were established between the indexes, including degree of hydrolysis,and the factors that affect the preparation of almond peptides by orthogonal rotation design. base on it, papain was chose to hydrolyze almond protein.According to experiments,The factors were optimized: temperature 50℃, substrateconcentration2, enzyme concentration [E]/[S]%4%, time 180min.on this condition almond protein's degree of hydrolysis for 27.22%. At best enzymolysis processing condition, through the hydrolysis of actual experiment get best yield for 28.02%.The desalting method by cation exchange resins and anion exchange resins mixed bed was built to improve purity of almond peptides. The optimum conditions were obtained: hydrolyte flow rate 10BV/h, pH value of hydrolyte 4.5 and between anion exchange resins and cation exchange resins 3:2, while the desaling method. Almond peptide recovery rate were all above 80%, the effect was better than traditional desalting method. For another, by the experiment of SDS- PAGE electrophoresis, the molecular weight distribution was below 2000Da.In this paper, active peptide can be prepared out by experiments, peptide recovery rate is 76.65%.also provided accodings for industrialization.
