Piceid is the main active ingredient of Polygonum cuspidatum, which has beenextracted mainly from the wild Polygonum cuspidatum, and caused the severe shortageof wild resources because of the long-term excavation. Therefore, studies on theextraction and separation of cultivated Polygonum cuspidatum as well as the detectionof the relative activities of piceid have important theoretical and application values. Inthis study, the cultivated Polygonum cuspidatum was taken as materials, it wasestablished of the method of piceid detection and determined of the content variationrules of piceid and other active ingredients in cultivated Polygonum cuspidatum. It wasalso studied of the best conditions of piceid extraction and separation. Meanwhile it wasexplored of the anti-aging activities of piceid through C.elegans model.1. Study on the establishment of piceid detection and its stability by highperformance liquid chromatography (HPLC). In this study, it was opimized of thedetection conditions of piceid, which were Diamonsil-C18250mm×0.46mm,5μmcolumn, the excitation wavelength334nm and emission wavelength408nm,with liquidflow1.0mL/min rate of acetonitrile (25%), and the temperature and injection volumewere25℃and10μL respectively. The results of the piceid stability study showed thatthe direct sunlight had great damage to the actiivities of piceid, while the impact ofindoor incandescent light was smaller, piceids was instability at high temperatures (>70℃).2. Detection of the content variation rules of piceid and other active ingredients incultivated Polygonum cuspidatum. In this study, the content comparisons were made ofresveratrol, emodin, emodin monomethyl ether, rhein and the other substances extractedin different years, different months, and different parts of cultivated Polygonumcuspidatum, the results showed that the contents of both the piceid and resveratrol ofcultivated Polygonum cuspidatum grew in August were the highest, while theanthraquinones such as the emodin and others were higher from July to September, andthe active ingredient contens were increasing year by year, meanwhile it was discoveredthat the active ingredient contents in the roots were higher than that in the stems andleaves.3. Study on the extriaction and seperation of piceid in cultivated Polygonumcuspidatum. The piceid was extracted by ethanol extraction method andmicrowave-assisted extraction method.Meanwhile the comparisons were made of theextracion results of these tow methods, which suggested that ethanol extraction wasoptimal under the conditions of65%ethanol as solvent, extracted for3hrs, that theamount of piceid reached to13.83mg/g. Additionally LSD-001was selected for piceidseparation, by which the content of piceid extraction raised from9.3%to51.4%, with the recrystallized purity of91.5%.4. Anti-aging activity of piceid. By the Caenorhabditis elegans model, it wasdiscovered that under the condition of the concentration of50mg/L of piceid, thelifespan of C. elegans in treated group was significantly longer than the control group,which increased by13.07%. The experimental results further showed that theswallowing rate, motility, and intestinal lipofuscinosis, the reproductive capacity ofpiceids in treated group were significantly enhanced than the control group, and thelipofuscin reduced was also observed in experimental group, which suggested that thepiceid extracted from cultivated Polygonum cuspidatum had more efficient anti-agingactivities.
