Objective: Epilepsy is a common chronic disease of the nervoussystem.It is one of syndromes that cerebral nervou sudden dischargeabnormally results in short-term cerebral dysfunction,characterized bysudden and recurrent.In our country there is about6-7million peoplewith epilepsy,part of these people are women of childbearing age.It isabout0.3%-0.4%of the newborns from mothers with epilepsy each year.2%-6%of children delivered by epileptic mothers have developmentdelay.At the samen time,the probability of mental disorders is2-7timesof the children delivered by the normal mother.Hippocampus play animportant role in the cognitive function such as learning and memory.andit is more vulnerable to be impaired than other brain regions facing thestress and orther harmful stimulate.The cognitive dysfunction of theoffspring is more and more to attention.Therefore,it gets more concernthat the influence of embryo development that PTZ-inducedpregnancy rat seizuring.In our study,by intraperitoneal injection ofpentylenetetrazol (PTZ) kindle the female Sprague-Dawley (SD)rats.Make the kindled female rat and male rat in a cage.Pregnant ratscontinue to injecting pentylenetetrazol by intraperitoneal.On thepregnant20day,we observe the live births, stillbirths number, absorbedfetus, fetal rat weight, length and staining fetal rat bone with Alizarinred to observe bone development.Detect the expression of bax, bcl-2andcaspase-3protein in hippocampal.Methods:We establish chronic epileptic model inducing bypentylenetetrazol (PTZ) intraperitoneal injection.30female SD rats(7-8weeks, body weight200±20g) were randomly divided into normal control group (NC group), saline group (NS group) and the epilepsygroup (PTZ group,).and every group has10rats.Since the first day,Therat in the PTZ group every day at9:00were injected PTZintraperitoneally by35mg/kg, the rat in NS group were intraperitoneallyinjected with the same doses of saline, the NC group received notreatment.According to Racine criterion, if we observed three timessuccessive seizures of grade Ⅳor V,the lighting in epileptic rats wassuccessful.After one week of the female rat kindled,female rat and malerat were co-cage,in the next day morning,we obsere vaginal plug,if therehas vaginal plug,we will make the day as0days of gestation.And everygroup continue to pre-pregnancy treatment until20days pregnant. Ineach group we would selecte randomly6rats,and take out the the fetalrat by caesarean section. Observe the number of live births in the fetal rat,the number of stillbirths, absorbed fetus, weigh up body weight, measurebody length, stain fetal rat bone with alizarin red to observe bonedeformity, and the ossification points of tailbone, sternum and spine. Theremaining pregnant rats take out the the fetal rat by caesareansection.The expression of caspase-3,bax and bcl-2protein in hippocampuswas determined through the methods of Western Blot.When the rat diebecause of seizuring,we would make a new rat to this groupResults:1Results of fetal mice morphology:Pregnant20days bycaesarean section get the fetal mice,Comparisons were made among theproportion of live births,,the proportion in PTZ goup is much lowerthan that in NC and NS goup(P<0.05);There was no significant differencebetween NC group and NS group (P>0.05). Two abnomal rats wereobserved in PTZ group,none was found in NC and NSgroup,however,There was no significant difference among the threegroup.Compare with the weight and the body length of the livefetal,There was no significant difference among NC group NS groupand PTZ group(P>0.05).2Resluts of fetal rat bone staining:There were no obvious skeletal deformities in the NC group, NS group and PTZ group rat.No significantdifference was found in the development of the supraoccipital boneand the sternum(P>0.05).The rate of bipartite ossification of vertebralcentrum in PTZ group is higher than NC group and NSgroup(P<0.05),but there was no significant deference between NC groupand NS group(P>0.05).3Results of Western blot:The expression of caspase-3protein inPTZ group was increased obviously compared to NC group(P<0.05).There was no significant deference caspase-3expression betweenNC group and NS group(P>0.05).According to expression of baxprotein,there was the same result.The expression of bcl-2protein in PTZgroup was decreased obviously compared to NC group (P<0.05).Therewas no significant deference bcl-2expression between NC group and NSgroup(P>0.05).Conclusions:1Seizuring during pregnancy may impact less onoffspring, and may delay the development of bone.2Seizuring during pregnancy make the expression of pro-apoptoticprotein bax increased and the antiapoptotic protein bcl-2reduced,at thesame time,the expression of apoptosis execution protein Caspase-3increased, speculate that mother with epilepsy seizures during theirpranancy make the hippocampal neuron apoptosis increased.
