Flavonoids and allaloids in mulberry leaves extract are widely applyied in pharmacy and food industry because of its function of hypoglycem, at present, the breed and specification of mulberry leaves extract are various, and they affect the move and development of market, establishing an quality standard of mulberry leaves extract can solve the problem completely.In this paper, mulberry leaves extract rich in flavonoids and alkaloids was chose as the object, and studied the determination, synchronized extraction process of flavonoids and DNJ, and make the quality standard of the mulberry leaves extract, then study the purification process of flavonoids.(1) A high performance liquid chromatography (HPLC) analysis method to determine flavonoids in mulberry leaves extract has been developed, and optimize the hydrolytic condition of flavonoid glycosides, the determination was so steady and reliable that totally satisfy the detection of flavonoids in mulberry leaves.(2) A synchronized efficient extraction process of flavonoids and 1-deoxynojirimycin(DNJ) were studied, The optimum extraction conditions of both flavonoids and DNJ were the liquid to solid 10:1, the ethanol volume 60%, pH=2,the extracting time 2h,the extracting temperature 80℃, 2 times. The extraction rate of flavonoids and DNJ were 82.4 % and 78.6 %.(3) The purification conditions of flavonoids was studied, the optimum conditions were: the feed solution concentration 2.93 mg/mL, pH 4, the flow rate 2 BV/h, adsorption volume 4 BV. Distilled water was washed for 2 BV, flow rate 2 BV/h. 70% alcohol as eluant for desorption, the flow rate 1 BV/h, and eluant volume 3 BV, obtained purificatory flavonoids after distilled and dried, the content was 212.69 mg/g.(4) The quality standard has been established: The content of flavonoids and DNJ in Mulberry Leaves extract were 50 mg/g and 1.5 mg/g.
