Objective:To study the effect of capsaicin and phenol on model of mouse allergic contact dermatitis,to investigate the relationship between nervous system and allergic contact dermatitis and approach the potential mechanism.Methods:48 BALB/c male mouse of 6-8 week old,clean grade,were randomly divided into 12 groups.We established the allergic contact dermatitis mouse model by topical application of 2,4-dinitrochlobenzene on the abdominal of the mouse to sensitize,then on the ear to stimulate. Several parameters were used to evaluate the severity of ear alterations,including ear swelling,diference of bilateral ear weight,ear thickness measured under the microscope,infiltrative cells in the tissue,and cell proliferation in the local lymph nodes. In the first experiment,we pretreated with specific neurotoxin:capsaicin or general neurotoxin phenol on mouse before the induction stages or provocation stages,and abserved the effect of capsaicin and phenol on the induction stages (afference ramus) or provocation stages (efference ramus) of mouse allergic contact dermatitis.In the second experiment,we set the interval as 3 ,6 ,9 ,12 and 15 days,deactivated cutaneous nerve fibres with capsaicin 2 days before sensitization,and abserved the relationship between the effect of capsaicin on mouse allergic contact dermatitis and time.Results:1. The mouse model of allergic contact dermatitis established successfully.In the model group and all control groups, bilateral ear swelling, ear weight, ear thickness measured under the microscope, infiltrative cells in the tissue, and cell proliferation in the local lymph nodes of the mouse had statistical difference(p<0.01).2. When cutaneous nerve fibres were deactivated by capsaicin or phenol or all peptide nerve fibres were deactivated before sensitization,all indexes between bilateral ears had no statistical difference(p>0.05). When cutaneous nerve fibres were deactivated by:capsaicin before provocation,all indexes between bilateral ears had no statistical difference(p>0.05).3. After the mouse were pretreated with capsaicin 2 days before sensitization,we set the interval between induction stages and provocation stages as 3 ,6 ,9 ,12 and 15 days,.All indexes between bilateral ears had no statistical difference(p>0.05).Conclusion:1. Capsaicin prevent the establishment of allergic contact dermatitis mouse model by preventing the afference ramus (induction stages) and efference ramus (provocation stages) of mouse allergic contact dermatitis.2. Phenol prevent the establishment ofallergic contact dermatitis mouse model by preventing the afference ramus (induction stages) of mouse allergic contact dermatitis.3. Nervous system plays an important role in the establishment and development of mouse allergic contact dermatitis.
