OBJECTIVES: (1) To study the suppressive effect of Chuankezhi (CKZ) injection on murine allergic contact dermatitis (ACD) induced by 2,4-dinitrofluorobenzene (DNFB) for three times. (2) To investigate the mechanism that CKZ suppress type IV hypersensitivity reaction through detecting the levels of IFN-γ, IL-2, TNF-a, IL-4, IL-10 in murine serum .METHODS: (1) Mice were divided into 6 groups according to different drugs treatments: they were CKZ high ,middle ,low dose (CKZ I II III) groups, Dexamethasone (DEX) , benadryl and normal saline (NS) groups. (2) Murine ACD induced by DNFB for three times were elicited. (3) Different drugs were administrated by intraperitoneal injection at 2h before sensitization; on day 1, day 2; 2h prior to and 6h following each challenge; on day 13 and day 14. (4) Effects of various drugs on murine left ear swelling, left ear flake weight, the count of dermal inflammatory infiltration cells and body weight were observed. Murine surum levels of Th1 type cytokines (IFN-γ, IL-2, TNF-α) and Th2 type cytokines(IL-4, IL-10) were measured by Luminex~?100 after challenge.RESULTS: The results showed that CKZ Ⅰ,Ⅱ,Ⅲ and DEX significantly inhibited the increase of left ear thickness, left ear flake weight and the count of dermal inflammatory infiltration cells compared with NS (P<0.05). Although DEX had a little more effectively suppressive effects (P<0.01), body weight difference in DEX group was significantly differrent compared with that in NS group (P<0.05). CKZ in three doses significantly suppressed upregulation of IFN-y(P<0.01) and DEX statistically suppressed upregulation of both IFN-γand IL-2 (P<0.01) in murine serum following challenge compared with NS.CONCLUSIONS: CKZ can significantly suppress murine ACD induced by DNFB for three times, CKZ can significantly suppress upregulation of IFN- v following challenge of ACD .which may be one of important mechanism that CKZ suppress type IV hypersensitivity reaction.
