Objective:To study PHT in human periodontal ligament cells express when it was stressed force,discuss the using tissue compatibility silicone membrane cultivate are on proligeration inhibition,PHT could make a role in promoting proliferation.To peovide a new idea for further research on periodontal damaged solution,and provide a new idea for periodontal trauma healing and tissue regeneration function provides experimental data support by PHT.Methods:Choose the normal premolar eradicated because of the orthodontic treatment, using tissue mass of training on the method of periodontal membrane cells. Configurat 20,50,100,500,2500ug/ml PHT cultures of different concentration gradien. Using tissue compatibility silicone membrane cultivate human periodontal ligament cells. Choose multimode biological tissue of silica gel film test equipment force stressing. Applying MTT method to detect 20,50,100,500,2500ug/ml PHT,15% stress force with 4 hours and after stressing force with certain concentration PHT to test the human periodontal ligament cells proliferation.With SPSS software for the single factor analysis of variance.Results1. The organization block stick wall of test results:wave silk protein positive, fight keratin negative.2. In the PHT concentration of 20,50, 100ug/ml, cells form no obvious change; when the concentration of 500,2500ug/ml, the cells are obvious toxic effects, the cellular structure damage, present a crystalline substances. With the 500ug/ml, there are a small group of stick wall cells; 2500ug/ml, no stick wall cells appear. After the MTT join 4h terminated, DMSO intermittence,thare has content of formation galleries needle crystallizers.With the same result by MTT:20mg/ml concentration of PHT, human periodontal ligament cells could promote proliferation obviously.3.With the 10%,15%,20% stress force in 4 hours, cells stick wall well in microscope,with no obvious directional changes with compared.With the MTT show that human periodontal ligament cells could significantly promote with 10% stress force,and the blank group was significant difference(p<0.05).Under 15% and 20% stress force, the human periodontal ligament cells proliferation of strain by significantly inhibited with blank group,with the significant difference(p<0.05).15% and 20% stress force no significant difference between them(p<0.05).4.20 ug/ml PHT can obviously reverse the result of excessive stress force due to the hPDLC proliferation inhibit.Put the human periodontal ligament cells with 15% stress force in 4 hours join 20mg/ml PHT to continue to cultivate 20h, by MTT detection, cell proliferat obvious, and the blank group has significant difference (p< 0.05), and 15% stress strain 4h group was significant difference (p< 0.01).Conclusion:1.Different concentrations of PHT periodontal ligament cell proliferation have significant differences.2.The periodontal ligament strain on appropriate stress force can promote periodontal cell proliferation,and with excessive stress force, cells produces proliferation inhibition.3.Proper concentration PHT could reverse the periodontal ligament cell proliferation inhibiting further promoting.
