Drug resistance is closely related to tuberculosis outbreak again from 1980s and prensents distinct characteristics in different area and country. Due to the great population movement, the tuberculosis is prevelent currently and the drug-resistance rate is increasing in Suzhou. It is very important for prevention and cure of tuberculosis in Suzhou to analyze the characteristics of katG and rpoB gene mutations of mycobacterium tuberculosis corresponding with isonaizd and rifampicin resistance.The epidemiological surve on drug resistance in tubdrculosis cases from September 2008 to May 2011 in Suzhou was performed by statistical analysis. The results presents as follows: 1. The total drug-resistance rate of four drugs INH, RFP, SM and EMB is 33.8% and each kind of drug-resistance rate is respectively 24.6 %, 13.3%, 24.7% and 10.7% for INH, RFP, SM and EMB, which are all higher than the average value of the world form 1997 to 2004. 2. The drug-resistance rate of HRSE and RHSE combination is respectively 22.7% and 40.0%, which is respectively higher than the single resistant rate of INH or RFP. 3. The total number of tuberculosis increased form September 2008 to May 2011 in Suzhou. The total resistant rate, each kind of the resistant rate of INH, RFP, SM and EMB and the MDR-TB oberviously increased and all of them present statistical significance. At the same time, the drug-resistant rate of the combination of four kinds of drugs increased form September 2008 to May 2011 and presents statistical significance.The mutation characterists of katG geng from MTB in Suzhou was investigated by extracting genomic DNA, PCR and sequence identification. The results show that A234G, S315T and R463L are the main forms of katG mutation and among of them S315T mutation is significant corresponding with INH resistant. While S315T and R463L present no statistical significance.Subsequently the mutation characterists of rpoB geng from MTB in Suzhou was also investigated by extracting genomic DNA, PCR and sequence identification. The results show that the typical mutation in RRDR of rpoB gene is S531L, the incidence rate of which is 60.5 percentage in RFP resistant Mycobacterium tuberculosis.
