| Objective: To investigate the expression level of MCT1 in rat cardiac myocardial cells and fibroblasts during ischemia and reperfusion.Methods: Single myocardial cells and fibroblasts isolated from rat hearts were subjected to the conditions mimicking ischemia and reperfusion. Ischemic condition was produced by culturing myocardial cells and fibroblasts with hypoxic substrate-low glucose solutions for 3 hours. To detect supernatants lactate concentration and use western blot and PT-PCT assays to determine the expression level of MCT1 at four time point: before ischemia(C), after 3h ischemia (I), after 30min reperfusion(R1) and after 60min reperfusion (R2).Results:1. MCT1 was expressed both in rat cardiac myocardial cells and fibroblasts,and its expression level was obviously higher in myocardial cells (P<0.01).2. The expression level of MCT1 in myocardial cells decreased after 3 h ischemia increased during 30min reperfusion after ischemia and was normalized after 60min reperfusion.3. The expression level of MCT1 in fibroblasts was increased during ischemia and reperfusion (P<0.01).4. The supernatants lactate concentration was decreased during ischemia and reperfusion (P<0.01).Conclusion: In conclusion, MCT1 is expressed both in rat cardiac myocardial cells and fibroblasts. We show for the first time that MCT1 expression is different between myocardial cells and fibroblasts. The MCT1 expression is increased in myocardial cells at 30min reperfusion after 3h ischemia, and increased in fibroblasts after 3h ischemia. Furthermore, the supernatants lactate concentration may associate with the expression of MCT1, and myocardial ischemia reperfusion injury may relate with the disorder of lactate transport between myocardial cells and fibroblasts. |
