Purpose:1.This study was designed to delineate the clinical implications of insulin-Like growth factor type 1receptor,growth factor receptor,Vascular endothelial growth factor and Human epidermal growth factor receptor 2 expressions in GC.2.To investigate the relationship between the features(sex,age, diseased region , Borrmann types, differentiation levels,TNM staging , Chemotherapy Regimen IGF-1R, EGFR, VEGF, HER2 , and prognosis of gastric cancer.METHODS:1.Clinical data of 108 Patients with gastric cancer who be in Guangzhou Medical College Affiliated Tumor Hospital between January 2003 and December of 2005.2.Using immunohistochemical techniques, we analyzed the expressions of IGF-IR, EGFR, VEGF, and HER2 on formalin-fixed paraffin-embedded specimens of surgically removed primary tumors, investigated the relationship between them.3.Select sex,age, diseased region , Borrmann types, differentiation levels, TNM staging , Chemotherapy Regimen,IGF-1R,EGFR,VEGF,HER2 as observed factors, investigated the relationship between them and prognosis of GC. Statistical caleculations were Performed using Statistical Package for Social Science software SPSS13.0. Results:1.IGF-1R,EGFR,VEGF,HER2 expression and relevance in GC:(1) IGF-IR expression was found in 67 tumors(62%), EGFR expression in 55(51%), VEGF expression in 46(44%), and HER2 expression in 22 (20%). (2) Positive coexpression of IGF-IR and EGFR was found 39 tumors (36%), that of IGF-1R and VEGF in 34 tumors (31%), that of IGF-IR and HER2 in 22 tumors (20%). (3) Positive coexpression of IGF-IR,EGFR and VEGF was found in 16 tumors (15%), that of IGF-IR,EGFR and HER2 in 10 tumors (9%),that of IGF-IR,VEGF and HER2 in tumors 10(9%), that of EGFR,VEGF and HER2 in 10 tumors (9%). (4) Positive coexpression of IGF-IR,EGFR,VEGF and HER2 was found 3 in tumors(2%).2.There are significantly positive corralationship between IGF-1R and EGFR,VEGF,HER2 expression in GC, R=0.252,0.233,0.356,P<0.05.3.Borrmann types, differentiation levels,TNM staging, Chemotherapy Regimen, IGF-1R, HER2 , IGF-1R and EGFR, IGF-1R and VEGF, IGF-1R and HER2, IGF-1R,EGFR and VEGF, IGF-1R,EGFR and HER2,IGF-1R,VEGF and HER2, EGFR,VEGF and HER2, IGF-1R,EGFR,VEGF and HER2 were influencing factors about Prognosis of gastric cancer.4.Multivariate survival analysis showed that PTNM(hazard ratio 2.185,95% confidence interval 1.623-2.941; P < 0.001),IGF-1R (hazard ratio 1.755,95% confidence interval 0.667-4.620; P = 0.0255),Borrmann types(hazard ratio 1.174,,95% confidence interval 0.684-2.013; P = 0.0261) were significant predictors of poor survival.Conclusions:1. Positive expression rate of IGF-IR significantly higher than the other three factors.2. there are significantly positive correlationship between IGF-1R and EGFR,VEGF,HER2 expression in gastric cancer.3. Borrmann types, differentiation levels,TNM staging, Chemotherapy Regimen, IGF-1R, HER2 , IGF-1R and EGFR, IGF-1R and VEGF, IGF-1R and HER2, IGF-1R, EGFR and VEGF, IGF-1R,EGFR and HER2,IGF-1R,VEGF and HER2, EGFR,VEGF and HER2, IGF-1R,EGFR,VEGF and HER2 were influencing factors about prognosis of gastric cancer.4. IGF-IR expression in surgical GC specimens, TNM staging, and Borrmann type tumors are significant predictors of poor outcomes in patients with GC.
